阿诺宁乳化剂的抗瘤作用和急性毒性试验
Studies on antitumor effects and acute toxicity of anuoning emulsion
【摘要】 目的 研究阿诺宁乳化剂的抗瘤作用和急性毒性反应。方法 用小鼠和裸鼠移植性肿瘤模型测定体内抗瘤活性 ,用Blliss方法计算小鼠给药的急性毒性的LD50 。结果 阿诺宁乳化剂在 4 ,8和 16mg·kg-1,灌胃 (ig) qd× 10d ,对小鼠肝癌HepS 3批实验的平均抑瘤率分别为 39.2 % ,4 6 .9%和 5 5 .7% ;对小鼠肉瘤S 180的平均抑瘤率分别为 34.6 % ,4 7.3%和5 4 .4 % ;对小鼠L2 的平均抑瘤率分别为 37.5 % ,4 5 .9%和 5 6 .5 %。阿诺宁乳化剂在 7.5 ,15 ,30和 6 0 μg·kg-1腹腔注射 (ip)×10d剂量下 ,对小鼠肝癌 (HepS)实验的平均抑瘤率分别为 31.0 % ,4 2 .9% ,5 0 .2 %和 6 2 .4 % ;对小鼠肉瘤S 180的平均抑瘤率依次为 2 3.6 % ,39.4 % ,5 0 .9%和 6 1.2 % ;在 15 ,30和 6 0 μg·kg-1,ip ,每日 1次 ,每周 6次 ,共 4周的剂量下 ,对人肝癌 (Bel 74 0 2 )裸鼠移植瘤的抑瘤率分别为 32 .0 % ,5 0 .5 %和 6 0 .2 % ;在上述 3个剂量下 ,对人肺腺癌 (GLC 82 )裸鼠移植瘤的抑瘤率分别为 30 .3% ,4 1.9%和 5 6 .1%。结果表明 ,阿诺宁乳化剂ig ,ip对上述 5种肿瘤均有明显的抑制作用 ,且其抑瘤率与阿诺宁乳剂的剂量相关。用Bliss方法统计出阿诺宁乳化剂对小鼠ig一次的半数致死量 (LD50 )及其 95 %可信限为 74 .75 (5 8.6 7?
【Abstract】 OBJECTIVE To investigate the antitumor effect and acute toxicity of anuoning emulsion in mice.METHODS The models transplanted tumor in mice and nude mice were used.Blliss method was used to calculate the lethal dose of 50% (LD 50 ) of anuoning emulsion in mice.RESULTS Under the doses of 4,8 and 16 mg·kg -1 anuoning emulsion intragstic (ig),qd×10 d,the average tumor inhibition rates were 39.2%,46.9% and 55.7% respectively ( P <0.01),against mice tumor HepS.The average tumor inhibition rates were 34.6%,47.3% and 54.4% ( P <0.01) against S-180 sarcoma in mice.In L 2 tumor,the mean inhibition rates were 37.5%,45.9%,and 56.5% ( P <0.05~0.01),respectively.Under the doses of anuoning emulsion 7.5,15,30 and 60 μg·kg -1 ,intraperitoneal injection (ip) qd×10 d to mice tumor HepS,the mean inhibition rates were 31.0%,42.9%,50.2% and 62.4% ( P <0.05~0.01),respectively.But In S-180 sarcoma the mean inhibition rates were 23.6%,39.4%,50.9% and 61.2% ( P <0.05~0.01),respectively.Under the doses of anuoning emulsion 15,30 and 60 μg·kg -1 ,ip×6/w×4 w,the inhibitory rates were 32.0% ,50.5% and 60.2% ( P <0.01) against xenograft tumor of human liver carcinoma (Bel-7402) in nude mice.Under the same doses,ip×6/w×3 w,the inhibition rates were 30.3%,41.9% and 56.1%,respectively,against xenograft tumor of human lung adenocarcinoma (GLC-82) in nude mice.The results showed that anuoning emulsion possessed antitumor effect against above-mentioned 5 tumors in mice and nude mice after ip and ig administration.In single intragastic (ig) management to mice,LD 50 and its 95% confidence interval was 74.75 (58.669~95.26) mg·kg -1 .In single ip management to mice,LD 50 of anuoning emulsion was 1.12 ( 1.01~ 1.24) mg·kg -1 .In single intravenous (iv) administration LD 50 was 1.81 (1.61~2.02) mg·kg -1 .CONCLUSION The experiment results indicated that anuoning emulsion possessed obvious antitumor effects against various transplanted tumor in mice and nude mice.
