Genistein抑制兔晶体上皮细胞膜酪氨酸蛋白激酶及蛋白激酶C磷酸化的研究
Inhibitive Effect of Genistein on Signal Phosphorylation of TPK and PKC in Membrane of Rabbit Lens Epithelial Cells
【摘要】 目的 :研究 genistein对兔晶体上皮细胞增殖及胞膜酪氨酸蛋白激酶 (TPK)、蛋白激酶C(PKC)活性的影响 ,探索使用 genistein防治后发性白内障。 方法 :兔晶体上皮细胞培养 ,应用Casnetllie改良法检测不同浓度genistein及bFGF作用后 ,兔晶体上皮细胞膜TPK活性的变化 ;同位素掺入法检测 genistein及bFGF作用后 ,兔晶体上皮细胞胞膜及胞浆PKC活性的变化。结果 :不同浓度 genistein作用后兔晶体上皮细胞TPK活性均较对照组明显下降 ,genistein可抑制bFGF诱导的TPK活性升高 ,genistein对PKC活性无明显抑制作用 ,但可抑制bFGF诱导的PKC活性升高。结论 :genistein在体外可通过抑制生长因子激活的蛋白激酶活性升高而抑制兔晶体上皮细胞增殖
【Abstract】 Objective: Our aim was to assess the effect of genistein on tyrosine protein kinase and protein kinase C (PKC) activation of rabbit lens epithelial cells (RLEC). Methods: The RLEC was cultured in vitro. Isotope incorporation method was used to measure activities of tyrosine protein kinase and protein kinase C in cell membrane. Results: The activation of tyrosine protein kinase was significantly inhibited by genistein. Activation of PKC in cytoplasm of lens epithelial cells induced by bFGF could be decreased by genistein. Conclusion: Genistein may be effective for the prevention of after cataract.
