蛋白激酶C同工酶在初发糖尿病大鼠肾小球中的细胞定位及表达
Localization and Expression of Isoforms of Protein Kinase C in Glomerular Cells of Early Diabetic Rats
【摘要】 目的 :探讨蛋白激酶C(PKC)同工酶α,βⅠ ,βⅡ ,ε在正常及初发糖尿病 (DM )大鼠肾小球中的细胞定位及表达变化与糖尿病肾病 (DN)发生、发展的关系。方法 :腹腔注射链脲菌素 (STZ)制成DM大鼠模型 ,采用免疫组化S P方法检测PKC同工酶的表达。结果 :PKCα在正常大鼠肾小球系膜细胞、上皮细胞、内皮细胞中均有表达 ;PKCβⅠ ,βⅡ 在系膜细胞中表达 ;PKCε在系膜细胞及内皮细胞中表达。初发DM状态下 ,PKCα在系膜细胞和内皮细胞中表达上升 ;PKCβⅠ ,βⅡ 在系膜细胞中表达明显下降 ;PKCε在系膜细胞及内皮细胞中表达升高不显著。结论 :PKC各同工酶在正常大鼠肾小球内的细胞定位不同。高血糖对PKC各同工酶在肾小球内的细胞表达有着不同的调控效应。PKC各同工酶在初发DM大鼠肾脏病变中发挥不同作用
【Abstract】 Objective: Our aims were to investigate the cellular localization and altered expression of protein kinase C(isoforms α,β Ⅰ,β Ⅱ,ε) in glomerular cells of normal and diabetic rats and to evaluate the relation between the altered expression of protein kinase C(PKC) isoforms and the pathogenesis of diabetic nephropathy(DN). Methods:The animals were divided into control group and diabetic group randomly. Hyperglycemia was induced with streptozotocin (STZ,60 mg/kg) in the male Sprague-Dawley rats(weight 150 to 170 g). Diabetes was confirmed when blood glucose was more than 13.9 mmol/L for 2 days after STZ administration. Age-matched normal rats received saline (3 mg/kg, 0.9%) and served as control. The expressions of PKC isoforms in normal and diabetic glomerular cells were assessed by using method of immunohistochemistry. Results:Isoform α was found in the 3 major glomerular cell types in normal rats. Isoform β Ⅰand β Ⅱ were shown in mesangial cells within the glomerulus. Isoform ε was detected in glomerular mesangial cells and endothelial cells. In early diabetic rats, immunohistochemistry revealed a marked increase of expression of PKC α in glomerular mesangial cells and endothelial cells. Expressions of PKC β Ⅰ and β Ⅱ significantly decreased in the glomerular mesangial cells.The expression of PKCε did not significantly change. Conclusion:The localizations of PKC α,β Ⅰ,β Ⅱ,and ε in glomerular cells of normal and diabetic rats are different. The altered expressions of PKC isoforms are not similiar in glomerular cells of early diabetic rats. The effects of PKC isoforms on the pathogenesis of DN are different.
