SARS冠状病毒基因组及其编码蛋白生物信息学分析

【摘要】 目的 :理论分析严重急性呼吸道综合征 (SARS)冠状病毒基因组及其编码蛋白的分子生物学特征 ,为进一步研究SARS冠状病毒蛋白质功能提供信息资料。方法 :利用生物信息学序列对比分析软件、蛋白氨基酸序列分析软件及蛋白质结构预测软件对GenBank公布的SARS冠状病毒基因序列及其编码的蛋白氨基酸序列进行分析。结果 :SARS冠状病毒在基因结构上具有冠状病毒属的特征 ,而又与其他已知的冠状病毒编码序列存在明显的不同 ,与其他冠状病毒相比没有明显的基因重组、大片段插入或缺失的现象。来源于不同SARS冠状病毒株的基因序列变异速度较快。出现基因序列在不同区段同源性差异的现象。S蛋白氨基酸序列不存在与其他物种相同的连续性抗原决定簇序列。结论 :SARS冠状病毒与其他冠状病毒的变异程度基本相同。SARS冠状病毒可能是自然界已经存在或变异产生的新的病毒株。近期世界范围流行的SARS是同一来源的病原体引起的 ,认为S蛋白氨基酸序列是SARS冠状病毒的主要抗原。

【Abstract】 Objective: Our purpose was to provide information for studies on protein functions of severe acute respiratory syndrome coronavirus (SARS CV) by theoretically analyzing the molecular biological characteristics of SARS CV, especially its genome and encoding proteins. Methods: We used several soft wares, including alignment comparison, amino acid sequence analysis, and protein structure prediction, in the analysis of the gene sequence and protein structure of SARS CV. The gene sequence of SARS CV was obtained from the Gene Bank. Results: The SARS CV has characteristics of coronal virus in gene structure, but in gene sequence, it is significantly different from the known coronal viruses. Compared with other coronal viruses, the SARS CV has no obvious gene reorganization and insertion or deletion of large fragments.The gene sequence from different SARS CV variated quickly. Therefore, different parts in the sequence have different autoploidy. The amino acid sequence of S-protein of SARS CV holds no identical continuous epitope with other species, which can be considered as the main antigen of SARS CV. Conclusion: No difference was found between the variation degree of SARS CV and other influenza coronal viruses, which indicates that the SARS CV may have already existed or, be a new virus produced via variation. The presently epidemic SARS in different areas of the world were caused by the same pathogen.
