反相高效液相色谱法同时测定人血浆中的帕罗西汀和舍曲林

【摘要】 目的 :建立一种简单、快速的同时测定帕罗西汀和舍曲林血药浓度的反相高效液相色谱法。方法 :以阿米替林作为内标。色谱条件 :C18反相柱 ,流动相为 0 0 1mol/L磷酸二氢钠 (pH 2 5 0 ) 乙腈 (6 4∶36 ,V/V)溶液 ,流速为 1 0ml/min ,在紫外检测波长 2 10nm处进行检测。结果 :帕罗西汀、舍曲林及内标在 12min内完全分离 ,帕罗西汀的最低检测浓度为 0 0 10mg/L ,在 0 0 15~ 0 75 0mg/L时线性关系良好 ,r =0 9998。舍曲林的最低检测浓度为 0 0 0 75mg/L ,在 0 0 10~ 0 75 0mg/L时线性关系良好 ,r=0 9998。低、中、高浓度下的回收率、日间及日内精密度均符合方法学要求。结论 :该方法可用于帕罗西汀和舍曲林血药浓度的临床监护。

【Abstract】 A simple and rapid reversed phase high perfor mance liquid chromatographic method was developed for the simultaneous determination of paro xetine and sertraline.A tricyclic antidepressant,amitriptylin e was used as an internal standard. Paroxetine and sertraline were isolated by liquid phase ex trac tion. Separation was obtained using a reversed phase column with a mobile phase of 0 01 mol/L NaH 2 PO 4 (pH 2 50) CH 3 CN (64∶36, V/V ). The detection wavelength was 210 nm.RESULT: Paroxetine,sertraline and amintriptyline were sepa rated within 12 min. The lower limit of detection was 0 015 mg/L of paroxetine and 0 0075 mg/L of sertraline. No endogenous compounds were found to interfere. The intra and inte r assay precisions were less than 10%,respectively.CONCLUSION: This m ethod proved suitable for drug monitoring in clinical research.
