镰刀菌毒素丁烯酸内酯对大鼠的急性毒性效应

【摘要】 目的 观察大鼠丁烯酸内酯急性中毒效应特征。方法 大鼠经口 1次灌服丁烯酸内酯 193mg/kg ,分别于中毒 4h和 2 4h后 ,观察其血象变化、血液生化指标及血浆钙、钠、钾、氯离子浓度的变化。结果 大鼠丁烯酸内酯急性中毒 4h后 ,红细胞总数、血红蛋白含量及红细胞压积容量均比正常对照组显著增加 ,粒细胞百分率显著增高 ,而血液淋巴细胞百分率明显下降 ;血小板总数亦明显增高。中毒 2 4h后 ,其血象已恢复至正常 ,与正常对照组比 ,差异无显著性 ;而血浆总蛋白、血浆白蛋白含量、Ca2 + 、血浆天门冬氨酸氨基转移酶及碱性磷酸酶活性明显降低。不同中毒时间大鼠血浆钠、钾、氯离子浓度、尿素氮、肌肝、总胆固醇及乳酸脱氢酶与正常对照组比较 ,均无明显改变。结论 大鼠丁烯酸内酯急性中毒起效迅速 ,中毒 4h时主要表现为脱水及粒细胞的增高 ;而中毒 2 4h时主要表现为血浆蛋白质的减少 ,提示以肝脏损伤为主要变化。

【Abstract】 Objective To observe the characteristic effects of acute poisoning induced by fusarium mycotoxin butenolide in rats. Methods 21 adult female Wistar rats were allotted at random to 3 groups of 7 each as Group I for control,Group Ⅱ and Group Ⅲ for butenolide poisoning.Rats were given 193 mg·kg -1 butenolide in a single dose by oral administration.After 4h and 24h poisoning,the blood of rats was used for determinations of serum chemistry and hematology. Results In the rats of 4h poisoning by butenolide,the hematology values such as red blood cells,hemoglobin,hematocrit,granulocytes and platelets were obviously increased.However,the serum chemistry did not change except cholesterol increase.For the 24h poisoning rats,all the parameters of hematology values recovered to normal as the control group.The serum chemistry including total protein,albumin,activity of aspartate aminotransferase and alkaline phosphatase was decreased.The serum chemistry,such as sodium,potassium,chloride,urea nitrogen,creatinine,and activity of alanine aminotransferase and lactate dehydrogenase did not change in all 4h and 24h poisoning rats as compared with control. Conclusions The toxic effects in rats appeared immediately after exposure to butenolide,the main toxic effect in 4h poisoning was dehydration and granulocytes increase.For 24h poisonig rats by betenolide the main toxic effect was serum protein decrease suggesting the liver injuries.
