大豆异黄酮抗高脂型大鼠主动脉血管壁粥样硬化斑块形成研究
Soy Isoflavones Inhibits Atherosclerotic Plaque Formation in Aorta of Hypercholesterolemia Rats and Regulates Gene Expression of Inducible Nitric Oxide Synthase and Adhesion Molecules
【摘要】 目的 研究高含量的大豆异黄酮 (ISO)与高脂型动脉硬化 (AS)体内发生过程中iNOS基因表达之间的关系。方法 采用总异黄酮含量为 72 4%的大豆异黄酮作为受试物 ,设 3 0mg/kg、 90mg/kg、 2 70mg/kg 3个剂量组 ,同时设雌激素 (己烯雌酚 )对照组 ,与AS高脂模型组、正常饲料对照组 ,喂饲Wistar大鼠 ,随时观察对大鼠体征 (体重、脏体比值 )的影响 ,运用光学显微镜检测HE染色的主动脉壁横切面病理改变 ,采用RT -PCR、Western -blot法检测血管壁内iNOS的mRNA和蛋白表达 ,并运用生化法检测血清中NO含量以及NOS活力。结果 大豆异黄酮可以剂量 -效应地抑制高脂饲料诱导的大鼠主动脉血管壁病理变化 ,增加血液中NO和NOS活力 ,显著增进血管壁内iNOS的mRNA和蛋白表达 (与高脂模型对照组相比P <0 0 5 )。结论 高脂饲料可以诱导大鼠主动脉壁形成动脉粥样斑块 ,并降低血清中NO含量和NOS活力 ,减少iNOS基因在主动脉内的表达 ,大豆异黄酮可以减轻高脂饲料诱导的主动脉壁病理变化 ,增进血清中NO含量和NOS活力 ,促进iNOS基因在主动脉内的表达 ,且这种作用可能与植物雌激素作用无关 ,与抗氧化分子结构有关
【Abstract】 Objective To Study the effect of soy isoflavones inhibits atherosclerotic plaque formation in aorta vessel of rats fed high cholesterol and high fat diets and study if this action is related to gene expression of inducible nitric oxide synthase(iNOS)and adhesion molecules such as ICAM-1,VCAM-1 Methods High content of purified isoflavones(containing 724 mg/g total isoflavones,285mg/g genistein)purification was used to treat rats fed high cholesterol and high fat diets to attenuates the formation of atherosclerotic plaque 60 male Wistar rats weight 160±20 g were separated into 6 groups randomly by total cholesterol level and the experiment lasted 20 weeks 50 rats were fed with high cholesterol and high fat diet(86 5% normal diet+3 5% cholesterol+10% pig fat+1% hyocholic salt) Among them,10 rats were classified as a model of atherosclerosis(B group),three groups of 30 rats fed with isoflavones as following:30mg/kg bw(C group),90 mg/kg bw(D group),270 mg/kg bw(E group) The rest 10 rats fed 0 25mg/kg bw estrobene serving as estrogen control group(F group)just because soy isoflavones are regarded as phytoestrogen Another 10 rats fed normal diet were used as a normal group(A group) Aorta was taken after 20 weeks,HE staining,Western-blotting and immunohistochemistry were used in this experiment Results Soy isoflavones at 90 mg/kg bw can attenuated the formation of atherosclerotic plaques which were evaluated by pathological features shown from microscope Soy isoflavones can lower the expression of ICAM-1 and V VCAM-1 and up regulates iNOS gene expression in order to increases the level of NO in serum so as to inhibit the formation of atherosclerosis
