猪乙脑病毒WHe株的PrM-E、NS1、E-NS2A基因的克隆及序列测定
Cloning and sequencing of NS1、prM-E、E-NS1-NS2A genes of pig Japanese encephalitis virus(WHe)
【摘要】 本文通过RT_PCR扩增了猪乙型脑炎病毒WHe株主要抗原基因NS1(约 1.14kb)、prM_E(约 2 .1kb)、E_NS1_NS2A (约 3.0kb) ,并将NS1、E_NS1_NS2A、PrM_E基因克隆、测序。与Genbank发表的JEV基因序列进行序列分析 ,发现WHe株与其他 12种典型的JEV强毒株的同源性在 88.3%~ 99.4 %之间。WHe株与K94P0 5株的同源性最低 (88.3% ) ,与P3株的同源性最高 (99.4 % ) ,可认为WHe株是由P3株衍生而来的。在JEV基因组 5’端 389~ 3910的长 35 2 2nt的决定JEV抗原性的C_prM_E_NS1_NS2A区中 ,WHe株与P3株仅有 2 1个碱基不同 ,其中的 14个单碱基突变为同义突变 ,另外7个为错义突变 ,导致相应的氨基酸序列 (1173个 )中的 6个氨基酸发生了变异 ,其中的 5个氨基酸变异出现在含有各关键的抗体中和决定簇的E蛋白内 ,另一个位于NS1蛋白内。
【Abstract】 Based on the WHe strain of Japanese Encephalitis Virus(JEV) isolated by Hubei Academy of Agricultural Sciences, we cloned NS1, C_prM_E and E_NS1_NS2A by RT_PCR and sequenced it. Comparison of the WHe strain 5’ one_third RNA genome with those of the other 13 typical JEV isolates revealed high level of sequence identity, ranging from 88.3 %(K94P05) to 99.4 % (P3) for nucleotides. It could be considered that the WHe was the derivative of the JEV/P3 strain. Among the 3522 nt long JEV genome region from position 389 to 3910, a total of 21 nucleotide differences were found between the Whe strain and the P3 strain. However, only 7 of those 21 mutations were missense mutations which cause the correspondent 6 unique amino acid differences. Among them, five occurred in the genome region that encoded the E protein while the sixth in the NS1 region.
