猪胰岛与大鼠睾丸支持细胞共同移植对糖尿病大鼠胰岛生长的影响
The cotransplantation of islets and Sertoli ce lls treats rat diabetes induced by streptozotocin
【摘要】 目的 研究胰岛异种移植的方法。 方法 用SD大鼠睾丸支持细胞 (Sertoli细胞 )与新生猪胰岛样细胞团 (ICCs)共同培养后进行异种移植。 结果 对照组胰岛细胞存活率及存活时间分别为 (63 .6%± 4.2 %和 3 .4d± 1.2d) ,较共同培养组 (87.2 %± 2 .7%和 13 .5d± 4.6d)显著为低(P <0 .0 1) ;对照组大部分胰岛细胞超微结构破坏 ,而共同培养组胰岛细胞超微结构基本正常 ;对照组胰岛素 2 4h累积分泌量和对葡萄糖刺激的反应性下降 ,共同培养组胰岛素分泌量始终处于高水平状态 (P <0 .0 1)。 结论 猪胰岛细胞与Sertoli细胞共同培养移植可以促进胰岛生长 ,明显延长胰岛移植物的存活时间
【Abstract】 ObjectiveTo inquire the method of xenotransplant ation of islet.MethodsThe xenotransplantation was done by us ing the cocultured Sertoli cells of SD rats with islet like cell clusters (ICCs) of neonatal pig and then the growing response and the effect of transplantation of coculture were observeol.ResultsThe survival rate and ti me of islets in control group were 63.6%±4.2% and 3 4 d± 1 2 d respective ly which were more decreased significantly compared with the coculture group(P <0 01 and most of the cell structure in this group were broken compared with the coculture group. The acumulative excretion amount of islet for 24 hours and the cell response to glucose stimulation also decreased in contral group and th ose in the coculture group were maintained in a higher level (P<0.01).After xenotransplantation,the mean survival time of graft cells in control and cocult ure groups were 3.4 d±1.2 d and 13.5 d±4.6 d respectively (P<0.01). ConclusionThe coculture of islets with Sertoli cells can improve the growth of islet and prolong the survival time of graft obviously. The xenotr ansplantation can increase the survival time of islet grafting. The xenotranspla ntation of coculture is an ideal method of islet culturing in vitro and graf ting.
