胰岛素受体底物-4基因多态性与2型糖尿病的相关性研究

【摘要】 目的 研究北京地区汉族人群胰岛素受体底物 4 (IRS 4 )基因密码子 879c g多态性与 2型糖尿病的相关性。 方法 采用配偶对作病例 对照研究。用聚合酶链反应 (PCR) 限制性内切酶片段长度多态性 (RFLP)的方法检测IRS 4基因密码子 879c g多态性。 结果  ( 1)男性与女性间IRS 4基因密码子 879c g多态性分布特点不同 ,男性无杂合子。g等位基因频率为男性 6 1.8%,女性6 3 .9%。 ( 2 )此多态性位点的基因型频率和等位基因频率在 2型糖尿病组和对照组间差异无显著意义。 ( 3)在男性对照组中 ,可见gg基因型组胰岛 β细胞功能指数低于cc基因型组。  结论 北京地区汉族人群中IRS 4基因密码子 879c g多态性与 2型糖尿病无明显相关性。

【Abstract】 Objective To study the association of insulin receptor substrate-4 (IRS-4) gene codon 879 c/g polymorphism with type 2 diabetes and its intermediate phenotypes in Chinese population in Beijing. Methods The IRS-4 gene codon 879 c/g polymorphism was detected by polymerase chain reaction and appropriate restriction enzyme (PCR-RFLP) in 110 type 2 diabetic cases and 80 normal controls. Results (1) There was different genotype distribution of IRS-4 codon 879 c/g polymorphism between the male and female groups. No heterozygote was found in male. The frequence of allele g was 61.8% in male and 63.9% in female respectively. (2) The genotype frequency and allelic frequency of the polymorphism were not significantly different between diabetics and control groups. (3) The islet β cell function index was lower in the individuals carrying the gg genotype than those carrying the cc genotype in male control group. Conclusions The gg geneotype of human IRS-4 codon 879 c/g polymorphism alone has no association with type 2 diabetes in northern Chinese population.
