猪IFN-γ基因的克隆与序列分析

【摘要】 猪IFN γ具有强烈的抗病毒、抗肿瘤和免疫调节作用 ,作为生物药剂和疫苗佐剂具有广阔的应用前景。通过对猪PBMC刺激诱导 ,提取总RNA ,然后采用RT PCR技术成功克隆了猪IFN γ基因。序列分析表明 ,克隆到的基因序列与NCBIGenBank上登载的猪IFN γ基因序列完全一致。该基因的成功克隆为IFN γ的进一步研究和开发奠定了基础。

【Abstract】 Porcine interferon gamma(IFN γ), a kind of cytokine, has been confirmed to be a key molecular in anti virus, anti tumor and immunity regulating function. It produced by activating T cell and NK cell. As a biological product and vaccine adjuvant, IFN γ plays an important role in clinical therapy and disease prevention. After porcine PBMC was stimulated by LPS and PHA, porcine IFN γ gene was cloned by the technology of reverse transcription polymerase chain reaction (RT PCR). In comparision with NCBI GenBank data,the homologies of the nucleotide sequence is 100%?This has laid a foundation for the production and development of porcine IFN γ.
