不同脉冲频率的半导体激光对荷肉瘤180小鼠血清IL-2含量和肿瘤生长的影响
Effect of Different Pulse Frequency Diode Laser Irradiation on IL-2 of the Sarcoma 180 Tumor-bearing Mice
【摘要】 目的 了解脉冲半导体激光照射对荷肉瘤 180小鼠血清中IL 2含量的影响和对肿瘤的抑制作用。方法 实验采用波长 65 0nm脉冲半导体激光。 40只荷瘤小鼠随机分为 4组 ,3组为激光照射组 ,1组为对照组 ,每组 10只荷瘤小鼠。每天分别用 10 0、5 0 0和 2 5 0 0Hz等 3种不同频率的脉冲半导体激光照射荷瘤小鼠的脾区 ,每日 1次 ,共 10d。照射后第 10天 ,采用双抗体夹心ELISA法测定各组荷瘤小鼠血清中的IL 2含量 ,并称肿瘤重量 ,计算肿瘤抑制率。结果 3种不同频率脉冲半导体激光照射后 ,荷瘤小鼠血清中IL 2含量均有增加 ,荷瘤小鼠的肿瘤生长均受到抑制作用 ,3组间差异无显著意义。结论 一定剂量的脉冲半导体激光照射荷瘤小鼠可提高小鼠血清中IL 2含量 ,对肿瘤的生长产生抑制作用
【Abstract】 Objective To observe the antineoplastic effect of pulse wave diode laser irradiation on sarcoma 180 transplanted in mice and to find out the effect of pulse wave diode laser on IL-2 of serum. Methods S 180 Solid tumor models in mice were established and pulse wave diode laser with wavelength of 650 nm were used in the experiment. Forty tumor-bearing mice were randomly divided into four groups, three laser irradiation groups, and one control group, 10 tumor-bearing mice were in each group. The spleen area of tumor-bearing mice of three laser irradiation groups were separately irradiated by pulse wave diode laser with different laser pulse frequency (100 Hz, 500 Hz, 2 500 Hz), at an energy density of 156.6 J/cm 2. Then the contents of interleukin-2((IL-2) in serum of tumor-bearing mice were measured by ELISA method. The average tumor weight of each group was also measured, and the inhibition rates were calculated. Results The results showed the contents of IL-2 of serum in three laser irradiation groups were significantly increased compared with that in control one (P<0.01), and the average tumor weight in three laser irradiation groups were significantly decreased compared with that in control one (P<0.01). However the contents of IL-2 of serum and tumor weights were not different among three laser irradiation groups. The inhibition rates in the three laser irradiation groups were 48.5%, 42.6%, 36.4%, respectively. Conclusions The pulse wave diode laser irradiation on the spleen area of tumor-bearing mice could hinder the growth of the tumor, and the antineoplastic effect might be due to increase the contents of IL-2 of serum in tumor-bearing mice after irradiation by pulse wave diode laser at a certain laser dose and pulse frequency.
