N-乙酰-L-半胱氨酸对H2O2引起的血管内皮细胞损伤的拮抗效应
Effects of N-acetyl-L-cystein on endothelial cells injury induce by hydrogen peroxide
【摘要】 目的 观察N 乙酰 L 半胱氨酸 (NAC ,痰易净 )对H2 O2 引起的血管内皮细胞损伤的影响。方法 通过测定乳酸脱氢酶 (LDH)释放率和脂质过氧化物 (LPO)值 ,MTT法测定细胞增殖活性 ,流式细胞术测细胞凋亡率 ,研究体外培养的人脐静脉内皮细胞在不同浓度H2 O2 作用下的细胞损伤及凋亡 ,以及NAC对抗细胞损伤及凋亡的效应。结果 随着H2 O2 浓度增加 ,细胞凋亡率明显增高 ,细胞增殖活力降低 ,细胞脂质过氧化物生成增加 ,LDH释放率增加 ,并在一定范围内呈剂量 效应关系。在H2 O2 中同时加入 0 5mmol/LNAC ,可使细胞活力增强 ,凋亡率降低 ,细胞脂质过氧化物生成显著减少 ,LDH释放率降低。结论 NAC可以对抗H2 O2 引起的血管内皮细胞损伤。
【Abstract】 Objective The study was designed to investigate the protective effects of N acetyl L cystein(NAC) on human endothelial cells injured by hydrogen peroxide (H 2 O 2 ) Methods H 2 O 2 in sequential concentration was added into the cultured human umbilical vein endothelial cells (HUVEC) for 24 hours in serum free medium The proliferative activity of cells (MTT method),the releasing rate of LDH,the lipid peroxidation,and cell apoptosis were assayed.NAC of 0 5 mmol/L was added into the HUVEC co cultured with H 2O 2 for assessment of the antagonistic rate of NAC on H 2 O 2 .Results H 2O 2 increased the cell apoptosis,lipid peroxidation,LDH releasing,decreased the proliferative activity of cell in a H 2O 2 concentration dependent manner These effects of H 2O 2 were inhibited markedly by NAC Conclusion Our data indicated that NAC could prevent human endothelial cells from injury induced by H 2O 2 possibly through its regulation on GSH and GSSG production
