VA对小鼠胚胎中Hox-3.5表达的影响

【摘要】 目的 观察小鼠胚胎组织中Hox - 3 5mRNA的表达及VA缺乏和补充对其表达的影响。方法 初断乳的昆明小鼠 ,雌鼠 6 0只随机分为正常对照组 (N)、VA缺乏组 (A)、妊娠第 0d补充VA组 (B)和妊娠第 7d补充VA组(C)。N组饲含VA 4 0 0 0U/kg饲料 ,A组饲VA缺乏饲料 ,B、C组在交配前饲VA缺乏饲料 ,在交配后分别于妊娠第0d或妊娠第 7d饲含 10 0 0 0U/kg饲料。雄鼠饲以普通饲料。在妊娠第12d ,剖腹取出胎鼠 ,采用原位杂交方法探测小鼠胚胎Hox - 3 5mRNA的表达 ,同时检查孕鼠血清VA水平。结果 在正常情况下 ,小鼠妊娠第 12d胚胎组织Hox -3 5mRNA有表达 ,孕鼠血清VA缺乏时 ,妊娠第 12d胚胎组织Hox - 3 5mRNA的含量明显减少 ;妊娠第 0d补充VA ,Hox - 3 5mRNA的含量与N组无差异 ;妊娠第 7d补充 ,Hox - 3 5mRNA的含量虽比A组增加 ,但仍明显低于N组。结论 VA可能在转录水平调控Hox - 3 5的表达。

【Abstract】 Objective To investigate the effect of vitamin A (VA) deficiency on Hox-3 5 mRNA expression in mouse embryos.Methods Sixty Kunming female mice were divided into four groups:normal control group N 〔4?000?U VA/(kg·diet)〕,VA deficiency group A 〔0?IU VA/(kg·diet)〕mVA-supplement after 0?d.p.c (day post coitus),group B and VA-supplement after 7 d.p.c group C.〔B、C were fed 0?IU VA/(kg·diet)〕 before crossover,B was fed 10?000 IU VA/(kg·diet) after 0?d.p.c and C was fed 〔10?000 IU VA/(kg·diet)after 7d.p.c.〕.On the 12th day p.c.pregnant mice in every group were killed and the uterus were cut open to take out the mouse embryos immediately.Meanwhile,blood was kept from pregnant mice for the determination of serum VA. In situ hybridization was used to determine Hox-3\^5 mRNA expression in the mouse embryos by digxigenin-labeled cDNA probes.Results Hox-3\^5 mRNA expression in the embryos of group A and C decreased significantly compared with group N and B ( P <0 01 or P <0 05), and there was no difference between group B and group N.The contents in group C multiplied compared with group A,but still reduced compared with group N.Conclusion VA may regulate Hox gene expression on transcriptal level.
