人胃癌组织中基质金属蛋白酶7的基因表达及意义
MMP-7 gene expression in gastric cancer and its significance
【摘要】 目的 :研究基质金属蛋白酶 7基因 (MMP 7)在胃癌组织及不同分化程度的胃癌细胞中的表达情况 ,探讨其临床意义。方法 :用RT PCR及凝胶成像仪检测MMP 7mRNA表达 ,用改良的BoydenChamber检测肿瘤细胞的侵袭能力。结果 :①MMP 7mRNA在胃癌组织中表达的阳性率为 60 % ,正常组织中表达的阳性率为 1 1 % ,肿瘤组织MMP 7mRNA表达量 (MMP 7/ β actin光密度平均比值为 0 .62 7± 0 .2 76)显著地高于正常组织 (为 0 2 37±0 1 2 ) (P <0 .0 5) ,同时有淋巴结转移的癌组织表达增高更为显著 ;②分化程度低的MGC 80 2、KATOⅢ、SGC 790 1细胞株MMP 7mRNA表达强阳性 ,而分化程度高的MKN2 8细胞株表达弱阳性 ;③转染了MMP7 asODN的KATOⅢ穿过膜的细胞数 (1 5 .6± 1 .2 1 )明显低于对照组、PS sODN组及PS mODN组 (分别为 75 .4± 6 .1 6、53 .8± 7.32、58.4± 5 .87) (P <0 .0 5)。结论 :MMP 7基因在胃癌组织中表达明显升高 ;分化低的细胞表达强阳性 ,而分化高的细胞表达弱阳性 ;MMP7反义寡核苷酸可明显降低肿瘤细胞的侵袭能力。MMP7在肿瘤浸润与转移中起作用。
【Abstract】 Purpose:To study the expression of MMP 7 gene in gastric cancer tissue and different stomach cancer cell line and explore its clinical significance.Methods:Reverse transcription polymerase chain reaction (RT PCR) was used to detect the expression of MMP 7 mRNA in 35 gastric cancer samples and the surrounding normal tissues and stomach cancer cell line. The ability of the invasion was observed by Boyden Chamber invasion assay. Results:The expression rate of MMP 7 mRNA in 35 gastric cancer samples and the surrounding normal tissues was 60% and 11%, respectively. The expression level of MMP 7 gene in gastric cancer was much higher than that in the surrounding normal tissues[(0 627±0 276)vs(0 237±0 12), P <0 05] Furthermore, the extremely high expression level of MMP 7 mRNA was detected in tumors with metastasis. The expression level of MMP 7 gene was strongly positive in low differential cell line .It was weakly positive in highly differentiated cell line. The number of cells migrating through the membrane obviously decreased after PS asODN treatment (15.6(1.21 )compared with the control group and PS sODN and PS mODN (75.4±6.16,53.8±7.32,58.4±5.87 respectively)( P <0.05).Conclusions:The expression level of MMP 7 mRNA is high in stomach cancer , especially in tumors with metastasis. It is strongly positive in lowly differentiated cell line, and weakly positive in highly differentiated cell line. MMP 7asODN reduced significantly the invasion ability of the cancer cell. MMP 7 play a role in tumor invasion and metastasis. [
