尿激酶与藻酸双酯钠治疗兔脑梗死模型的实验性对比研究

【摘要】 目的 为进一步探讨尿激酶和藻酸双酯钠 (PSS)对脑梗死的治疗作用机制。方法 将 3 2只家兔分为 3组 ,先将实验兔制成高脂、高糖血症模型 ,再制成家兔脑梗死模型 ,然后分别给予尿激酶、PSS和尼莫地平 3种药物治疗 ,观察实验兔的临床变化 ,同时动态检测血浆内皮素 (ET- 1)、P-选择素 (P- selec)、血管内皮生长因子(VEGF)水平 ,并与对照组做了比较。结果 结果显示高脂、高糖血症兔模型可使血浆 ET- 1和 P- selec水平明显升高 (P<0 .0 1) ,梗死后 2 h ET- 1和 P- selec水平明显升高 (P<0 .0 5~ 0 .0 1) ,梗死后 72 h兔的临床症状基本恢复正常。治疗后尿激酶仅能使 P- selec水平下降 (P<0 .0 5 ) ,PSS组可使 ET- 1、P- selec和血脂水平一并下降 (P<0 .0 5~0 .0 1) ,尼莫地平组无变化 ,3种药物对 VEGF水平均无影响。结论 高脂、高糖血症兔模型的血浆 ET- 1和 P- selec水平升高 (P<0 .0 1) ,脑梗死后 2 h ET- 1和 P- selec水平明显升高 (P<0 .0 5~ 0 .0 1) ,尿激酶、PSS和尼莫地平与对照组比较 ,PSS组对血浆生物分子改变最为显著 (P<0 .0 5~ 0 .0 1)。

【Abstract】 Objective To explore the mechanism of effect in the rabbits model with cerebral infarction treated by Urokinase and PSS. Methods 32 rabbits have divided into 3 groups. The rabbits models with hyperglycemia and hyperlipoidemia had made at first,so than the model with cerebral infarction which was carried to the blood clotts injected into the internal carotid and than ligated it. After that,the treatment was given respective with Urokinase,PSS and Nimodiping. At the same time,the manifestation of rabbit models were observed and the concentration of plasma ET 1 and P selec and VEGF were measured,The value of inspected was comparied to that of the controled group. Results The results showed that the concentration of plasma ET 1 and P selec were elevated during the rabbit models with hyperglycemia and hyperlipoidemia( P <0.01). The level of ET 1 and P selec were prominence elevated after 2h after infarction( P <0.05~0.01). The whole symptoms of cerebral ischemia recovered after 72h infarction. The level of plasma P selec only reduced by Urokinase( P <0.05),and PSS might cut down the level of plasma of ET 1 and P selec and the capacity of blood lipid( P <0.05~0.01),but there are no any changer in the Nimodiping after treatment. It was no effect on the VEGF in 3 groups. Conclusion The level of plasma ET 1 and P selec elevated by hyperglycemia and hyperlipoidemia in the rabbit models( P <0.01),but which it was prominence elevated in level of ET 1 and P selec after 2h after infarction( P <0.05~0.01). The biomolecule changes was remarkable in PSS group which compared with the Urokinase,Nimodiping and the coutrast group( P <0.05~0.01).
