砷与氟单独及联合染毒对SGC-7901细胞基因组DNA损伤作用的SCGE法观察

【摘要】 目的 观察砷、氟及砷氟联合染毒对SGC 790 1细胞基因组DNA的损伤作用及特点。方法 砷、氟染毒剂量 10 μmol/L ,染毒后 4h应用单细胞凝胶电泳技术结合彗星图像分析系统检测DNA损伤。 结果 ①NaAsO2 组尾长和尾动量明显大于对照组 ,NaF组拖尾率显著高于对照组 ;②氟砷联合染毒组拖尾率、尾DNA含量、尾长和尾动量与对照组差异均有显著意义 ,但氟与砷引起的DNA损伤作用之间没有观察到交互作用。结论 NaAsO2 与NaF均可引起DNA损伤 ,但它们的损伤机理可能是不同的 ;氟与砷联合染毒仅表现为简单相加作用

【Abstract】 Objective To study the features of DNA damage induced by arsenic, fluoride and their combination in cultured SGC 7901 cells.Methods Single cell gel electrophoresis(SCGE) and Komet Image Analysis System (KIAS) were used to measure the DNA damage of the cells treated with 10 μmol/L sodium and/or sodium fluoride for 4 hours.Results ①The tail length and the tail moment of damaged DNA were significantly greater in the sodium arsenite group than that in the control group, the tailed cell percentage in the sodium fluoride group was significantly higher than that in the control group;②Such indicators as the tailed cell percentage, tail DNA,tail length and tail moment in the combined treatment group were significantly increased , however the interaction of DNA damage between arsenic and fluoride was not observed.Conclusions Both sodium arsenite and sodium fluoride could induce DNA damage, but their mechanism of inducing DNA damage may be different. And the DNA damage effects of sodium arsenite and sodium fluoride in the combination group were simply additive.
