预防非典型肺炎方剂对小鼠外周血液淋巴细胞DNA损伤的影响
Effect of Chinese meteria medica prescriptions against SARS on lymphocyte DNA damage in mice
【摘要】 目的 研究预防非典型肺炎方剂对小鼠外周血液淋巴细胞 DNA的损伤作用。方法 应用单细胞凝胶电泳方法测定 6个预防非典型肺炎方剂体内给药对小鼠外周血液淋巴细胞 DNA损伤的作用。结果 ig给予预防非典型肺炎方剂 、 、 (1/ 3临床等效剂量、临床等效剂量、3倍临床等效剂量× 3 d)对小鼠外周血液淋巴细胞 DNA有明显的损伤作用 ,并呈剂量依赖关系。预防非典型肺炎方剂 、 、 (1/ 3临床等效剂量、临床等效剂量、3倍临床等效剂量× 3 d)对小鼠外周血液淋巴细胞 DNA没有明显的损伤作用。结论 预防非典型肺炎方剂 、 、 具有潜在的遗传毒性
【Abstract】 Object To evaluate the genotoxicity of the prescriptions against severe acute respiratory syndrome (SARS) in mice lymphocytes DNA damage in mice. Methods Six prescriptions were ig administered to the mice for 3 days. The DNA damage was measured in mice lymphocytes by means of single cell gel electrophoresis assay (SCGE, or comet assay). Results Lymphocyte DNA damage was distinctly observed by administration of the prescriptions Ⅰ,Ⅴ and Ⅵ for 3 days at the doses of 1/3 to 3 times of the clinically equivalent dosage. There was no significant DNA damage by administration of the prescriptions Ⅱ, Ⅲ and Ⅳ for 3 days. Conclusion The prescriptions Ⅰ, Ⅴ and Ⅵ possess potential genotoxicity.
