枸杞多糖-4对下丘脑损伤性肥胖小鼠减肥作用的探讨
EFFECT OF LYCIUM BARBARUM POLYSACCHARIDE-4 ON MONOSODIUM GLUTAMATE INDUCED OBESITY IN MICE
【摘要】 目的 : 研究枸杞多糖 -4对下丘脑损伤性肥胖小鼠的减肥作用及其作用机制。方法 : 雄性下丘脑损伤性肥胖小鼠模型造型后分别以 2 0、40和 60 mg/ ( kg· d)的剂量连续灌胃 3 0d后 ,称体重 ,测量体长 ,眼球取血处死 ,称全身脂肪重量 ,计算李氏指数和脂肪指数 ,测定血脂水平 ,将脂肪组织固定、染色后 40 0倍显微镜下计数单个视野内脂肪细胞个数 ,测定脂肪组织中乙酰辅酶 A羧基酶 ( acetyl-Co A carboxylase,Acc) m RNA含量。结果 : 枸杞多糖 -4可显著降低下丘脑损伤肥胖小鼠的体重、李氏指数和脂肪指数 ;60 mg/ ( kg·d)枸杞多糖 -4可显著降低血清 TC,各剂量组的血清 TG含量均显著降低 ,2 0和 40 mg/ ( kg· d)的枸杞多糖 -4可显著提高血清 HDL-C含量 ;并可显著减小脂肪细胞大小和增加脂肪组织内 Acc m RNA的含量。结论 : 枸杞多糖 -4可通过调节机体的能量代谢达到降脂减肥的作用。
【Abstract】 Objective: To investigate the effect of Lycium barbarum polysaccharide 4 (LBP 4) on monosodium glutamate induced obesity in mice. Methods: LBP 4 was applied to monosodium glutamate induced obese male mice at a dose of 20?40 and 60 mg·kg -1 ·d -1 for 30 days, respectively. The body weight, Lee’s index, fat index, lipids in serum, the volume of fat cells and the content of acetyl CoA carboxylase (Acc) mRNA in fat tissue were determined. Results: LBP 4 could decrease body weight, Lee’s index, fat index, lipid in serum and the volume of fat cells, and increase the content of Acc mRNA in fat tissue. Conclusion: LBP 4 can adjust the gene expression of Acc in fat tissue of male mice and can decrease fat content in monosodium glutamate induced obese male mice.
