食用仙人掌‘米邦塔’的试管快繁
Rapid Propagation of Cactus‘Milpa Alta’in Vitro
【摘要】 对食用仙人掌‘米邦塔’的试管快繁研究结果表明 :初代培养基以MS + 6 BA 0 6mg·L-1 +NAA 1 0mg·L-1 + 2 ,4 D 0 1mg·L-1 为最佳处理 ,接种 6 0d后 ,外植体上刺座下的潜伏芽萌发率可达 6 7%。继代增殖培养适宜培养基为MS + 6 BA 0 1mg·L-1 +NAA 1 0mg·L-1 + 2 ,4 D 0 2mg·L-1 或MS + 6 BA 0 1mg·L-1 +NAA 0 5mg·L-1 ,平均每 50d继代增殖培养 1次 ,增殖倍数可达 5~ 6 ,且新生的小子茎生长旺盛。在培养过程中 ,虽然部分外植体切口处发生大团绿色至黄绿色小颗粒状愈伤组织 ,然而均没有分化不定芽。在生根培养基MS+IAA 0 1mg·L-1 或MS +IBA 0 1mg·L-1 上 ,2~ 3cm高的小子茎培养 10d左右发根 ,形成完整植株。当试管苗高达 5cm时移入砂床 ,成活率高达 99% ,经约 6 0d ,试管苗发出第一批掌片
【Abstract】 Effects of several factors such as medium,cutting mat erials and sterilization methods for rapid propagation in vitro on cactus(Opun tia dillenii)‘Milpa Alta’were studied.The results indicated that the optim al medium for primary culture was MS+6-BA 0 6mg·L -1 +NAA 1 0 mg·L -1 +2,4-D 0 1mg·L -1 . Taking c actus fresh stem as explant,a shooting rate of 67% of latent bud under the spine -base was investigated after 60 days of inoculation on this medium.The appr opriate medium for multiplication and subculture of shooting were MS+6-BA 0 1 mg·L -1 +NAA 1 0mg·L -1 +2,4-D 0 2mg·L -1 or MS+6-BA 0 1mg·L -1 +NAA 0 5mg·L -1 .On this medium shoot multiplication occured at a 5 to 6-fold rate after 50 days.The shoots cultured on the rooting medium of MS+IAA 0 1mg·L -1 or MS+IBA 0 1mg·L -1 produced rootlets after 10 days.The p lantlet upwards of 5cm height was rooted in sand medium with up to 99% s urvival and produced new shoots in the field after 60 days.
