苯巴比妥在患儿血清样品中的稳定性

【摘要】 目的 :探讨患儿苯巴比妥 (PB)血清样品在冷处储存的稳定性。方法 :荧光偏振免疫法 (FPIA)测定 3 0例患儿应用PB治疗时的血药浓度 ,比较其血清样品当天测定浓度 (C1 )与其在冷处储存 7d后测定浓度 (C7)。结果 :PB血药浓度C1 与C7呈正相关 (P <0 .0 1) ,其回归方程 :C7=1.0 5 45C1 -1.2 75 8,r=0 .983 1。平均Re(C7 C1 ,相当于PB在患儿血清中的平均回收率 )为 (10 0 .4± 4.8) % ,RSD为 4.70 % (n =3 0 ) ,且两者的各组偏差 [(C7-C1 ) C1 ]均小于 10 %。结论 :患儿血清样品在冷处储存 7d ,其PB能保持稳定 ,有利于常规治疗药物监测 (TDM)时的样品储存

【Abstract】 Objective:To probe into the stability of phenobarbital (PB) in refrigerated serum samples from sick children. Methods:The serum concentration of PB in 30 sick children treated with the drug was analyzed with the fluorescence polarization immunoassay (FPIA). The concentrations of PB in sera harvested on the day of blood sampling ( C 1) were compared with those in the sera which had been refrigerated at 2~10℃ for 7 days( C 7). Results:The blood concentration of PB C 1 was positively correlated with that of C 7 ( P <0.01), the formula of linear regression being C 7=1.054 5 C 1-1.275 8, r =0.983 1. The average Re C 7/ C 1, corresponding to the mean recovery rate of PB in sera of sick children, was (100.4±4.8)%, with an RSD of 4.70 ( n =30). Both the standard deviations [( C 7- C 1)/ C 1] were smaller than ±10%. Conclusion:The concentrations of PB in sera from sick children were shown to be kept stable after the sera had been refrigerated for 7 days. The results seem to be advantageous to routine therapeutic drug monitoring(TDM) for stored samples.
