吗啡依赖大鼠腹侧被盖区多巴胺神经元的形态学改变
MORPHOLOGICAL CHANGES OF VENTRAL TEGMENTAL AREA DA NEURONS IN MORPHINE WITHDRAWAL RATS
【摘要】 目的 :探讨吗啡依赖自然戒断时 ,中脑边缘多巴胺系统中腹侧被盖区 (ventraltegmentalarea ,VTA)多巴胺神经元的形态学改变。方法 :建立吗啡依赖大鼠自然戒断模型 ,用免疫组化方法对VTA多巴胺神经元着色 ,应用图像处理技术对神经元的形态学改变进行量化分析。结果 :(1)短期 (2d)戒断组神经元的光密度 (P <0 0 5 )、面积 (P <0 0 1)、周长 (P <0 0 1)、偏心率 (P <0 0 5 )、形态指数 (P <0 0 1)等参数指标较对照组发生了显著性改变 ;(2 )长期(14d)戒断组神经元的形态指数 (P <0 0 5 )较对照组发生了显著性改变 ;(3)短期戒断组的周长 (P <0 0 1)、偏心率(P <0 0 5 )、形态指数 (P <0 0 1)与长期戒断组相比有显著性差异。结论 :吗啡依赖自然戒断时 ,VTA多巴胺神经元发生了明显的适应性形态学改变 ,这种改变随戒断时间延长而逐渐弱化
【Abstract】 Objective: To study morphological changes of ventral tegmental area DA neurons in morphine natural withdrawal rats. Methods: Build up morphine natural withdrawal rats models, dye ventral tegmental area DA neurons by immunohistochemical method and analyze digital morphological changes of neurons by image processing techniques. Results: Compared with the normal saline (NS) treatment group, significant changes were observed in optical density (OD) (P<0.05), area (P<0.01), perimeter (P<0.05), eccentricity (P<0.05) and morphological index (MI) (P<0.01) of short-run (2 d) withdrawal group and in MI (P<0.05) of long-run (14 d) withdrawal group. There were significant differences in perimeter (P<0.01), eccentricity (P<0.05) and MI (P<0.01) between short-run and long-run groups. Conclusion: Significant morphological changes have taken place in ventral tegmental area DA neurons in morphine natural withdrawal rats, and the changes become weaker as time goes on.
