体外细胞株G0期同步化模型建立及分析
The development and analysis of cell synchronization model in G0 phrase
【摘要】 通过对U251,QGY,Hela3个细胞株进行血清剥夺培养,建立起有较好的同步化效果的U251同步化模型.细胞同步化后,转入血清培养基培养,在24h内分10个时间点收集血清恢复培养不同时刻的细胞样品,应用流式细胞仪测定周期进行情况.结果显示,在0.25和8h,有2个重要的G1/S转变点,提示血清剥夺造成的G0期同步不是由G1/S检查点所控制,而是另有值得深入研究的控制点.
【Abstract】 In order to develop a protocol of synchronize cell reversibly in the G0 phrase.Three cell line (U?251,QGY,Hela)were cultured in serum deprival medium,then they were switched to serum medium.Cell cycle were detected over 24 h with cell sample collected at 10 points.The fact indicated that the protocol is successful and the point of 0.25 and 8 h present two important regular point and cell G0 synchronization didn’t control by G1/S check point but another unknown point after serum deprival treatment.
