青海柯柯盐湖、茶卡盐湖细菌富集培养后DGGE带谱比较
Assessment of enrichment bacterial community in Keke and Chaka salt lakes of Qinghai by DGGE
【摘要】 分别将采自青海柯柯盐湖和茶卡盐湖的土样、泥样富集培养后,提取DNA,用细菌通用引物扩增得到含16SrDNA的V9高变区的DNA片断,进行DGGE(变性梯度凝胶电泳)分析.结果表明这2个相邻的盐湖中细菌多样性差异较大,同一盐湖中土样和泥样的细菌多样性也有较大的差异而且泥样中的多样性较土样的稍丰富些.
【Abstract】 Denaturing gradient gel electrophoresis (DGGE) was used to assess the enrichment bacterial community in two adjacent lakes,Keke salt lake and the Chaka salt lake, Qinghai,China.DNA was extracted followed by amplification of 16S rDNA,then PCR products which include the V9 highvariable region were analyzed by DGGE.The DGGE profiles showed that there are different bacterial communities in the two adjacent lakes and different bacterial communities exist in different samples of the same lake after enrichment,and the sludge samples show more diversity both in the two lakes.
