中国玉米新品种DNA指纹库建立系列研究 Ⅱ.适于玉米自交系和杂交种指纹图谱绘制的SSR核心引物的确定
Series of Research on Establishing DNA Fingerprinting Pool of Chinese New Maize Cultivars
【摘要】 本项研究利用60个玉米自交系对初选出的158个SSR引物对进行进一步筛选,综合考虑PIC值大小、扩增带型统计的难易及引物的重复性高低,确定bnlg439、bnlg125、phi053、bnlg197、phi072、bnlg238、phi126、bnlg161、bnlg240、bnlg619共10个引物对作为构建玉米自交系和杂交种指纹库的核心引物;同时提出适用于计算玉米SSR指纹图谱概率的公式:P=1/N,对自交系N=n;对杂交种,N=Cn1+Cn2。n为所用引物的等位基因数。
【Abstract】 The conception of core primer was defined.60maize inbred lines were used to screen158SSR prime pairs and finally a set of core prime pairs fitting for establishing maize inbred line and hybrid DNA fingerprint pool were confirmed,which were bnlg439,bnlg125,phi053,bnlg197,phi072,bnlg238,phi126,bnlg161,bnlg240and bnlg619.At the same time a formula for calculating existence probability of maize SSR fingerprint was advanced:P=1/N,for inbred lines,N=n;for hybrids,N=C n1 +C n2 .n is the allele number of the primer pairs.
