【摘要】 目的 通过激光诱导小鼠脉络膜新生血管(CNV)模型,探讨TGF-β1在CNV形成中的作用。方法 应用810nm半导体激光光凝C57 BL/6J小鼠眼底后极部,诱发CNV生成。术后1、4周,用原位杂交方法检测TGF-β1 mRNA在小鼠CNV中的表达。结果 在正常组织中,TGF-β1 mRNA表达于神经节细胞层,而视网膜色素上皮、脉络膜血管内皮细胞无TGF-β1的mRNA表达。激光光凝1周后CNV膜形成,TGF-β1 mRNA主要表达于CNV及其附近。激光后4周TGF-β1信号仍明显表达。结论 研究结果提示在CNV发生过程中,TGF-β1可能起重要作用。

【Abstract】 Objective To investigate the expression of transforming growth factor pi ( TGF-β1 ) in mice with choroidal neovascularization induced by diode laser. Methods Diode laser was applied to the posterior poles of the eyes in C57 BL/6J mice to induce choroidal neovascularization (CNV).The expression of TGF-β1 mRNA was detected by in situ hybridyzation in 1 and 4 weeks after photocoagulation. Results TGF-β1 mRNA was expressed in cells of ganglion cell layer, but was not detected in RPE layer and choriocapillaris in normal adult mice. TGF-β1 mRNA was mainly located in CNV in 1 week after laser treated, and was present even 4 weeks. Conclusion TGF-β1 may play an important role in the development of CNV.
