大鼠眼玻璃体内注射神经生长因子的药代动力学研究

【摘要】 目的 研究大鼠眼玻璃体内注射神经生长因子 (NGF)后的药代动力学特性。方法 SD大鼠 40只 ,随机分为 10组 ,每组 4只 8眼 ,任选一组SD大鼠作为正常对照组不注射NGF ,测正常玻璃体内的NGF质量浓度 ;其余各组SD大鼠均玻璃体内注射NGF 2 0 μg ,分别于注药后 5、15、3 0min ,1、2、4、8、12和 2 4h各处死一组大鼠 ,取出玻璃体样本 ,用间接酶联免疫吸附试验 (ELISA)测出玻璃体内NGF质量浓度。结果 正常大鼠玻璃体内NGF质量浓度为 3 69μg/L±3 75 μg/L。玻璃体内注射NGF后 ,在玻璃体内的消除半衰期 (t1/ 2 )为 0 93h( 5 5 8min) ,药 -时曲线下面积 (AUC0~t)为5 11 49μg/(L·h) 。结论 NGF在玻璃体内消除半衰期极短

【Abstract】 ObjectiveTo study the pharmacokinetics of nerve growth factor (NGF) after intravitreal injection in rat.MethodsFourty healthy SD rats were divided into 10 groups,of 4 animals each.In order to determine the concentration of NGF in normal rat vitreous,one group was randomly chosen without intravitreal injection.In other groups,NGF 20μg was injected into vitreous of each rat eye.Rats of each group were killed at 5,15,30min,1,2,4,8,12 and 24 hours after injection respectively and the vitreous specimens were obtained immediately.NGF concentrations were measured by the indirect enzyme linked immunosorbent assay (ELISA).ResultsThe concentration of NGF in normal rat vitreous was (3.69±3 75)μg/L.The half life for elimination (t 1/2 )of NGF from vitreous was 0 93 hours.The area under concentration time curve (AUC 0~t ) was 511 49(μg/L.h).ConclusionThe half life for elimination of NGF from vitreous is very short.Repeat administration of NGF is recommended to maintain an effective concentration.
