【摘要】 目的　研究MDM2 基因蛋白表达与大肠癌生物学行为及预后的关系。方法　应用免疫组化LDP法检测 6 0例大肠癌手术标本、30例癌旁组织及 16例大肠良性病变组织中MDM2 基因蛋白的表达。结果　大肠癌组织中 ,MDM2 基因蛋白表达阳性率为 75 % ( 4 5 /6 0 ) ,其中伴有区域淋巴结转移者阳性率为 95 .6 % ( 4 3/45 ) ,无伴有淋巴结转移者阳性率为 13 .3 % ( 2 /15 ) ,两者比较差异有高度显著性 ( χ2 =36 .30 ,P <0 .0 0 1) ;MDM2 基因蛋白表达水平在不同的大肠癌分化程度、浸润深度中差异有高度显著性 ( χ2 分别为 2 8.42 ,2 4.6 7,P均 <0 .0 0 1) ;但在不同的Duke分期、不同年龄组、不同部位及类型的大肠癌之间差异无显著性 (P >0 .0 5 ) ;MDM2 基因蛋白表达阳性者术后 5年生存率与阴性者比较差异有显著性 ( χ2 =6 .40 ,P <0 .0 5 )。 30例癌旁组织及 16例良性病变呈阴性表达。结论　MDM2 基因在大肠癌浸润和淋巴结转移过程中发挥重要作用 ,MDM2 基因蛋白的检测 ,可作为大肠癌预后判断的一个重要指标。更多还原

【Abstract】 Objective To study the relationship between MDM 2 gene protein expression and the biological behaviour and prognosis of carcinoma of large intestine.Methods An immunohistochemical LDP technique was used to detect MDM 2 gene protein expression in 60 surgical samples of carcinoma of large intestine, 30 lateral tissues of cancer and 16 benign lesion tissues of large intestine.Results In the tissues of carcinoma of large intestine, the positive expression rate of MDM 2 gene protein was 75% (45/60), of which the positive rate in tumors associated with regional lymphatic metastasis was 95.6% (43/45) and in tumors without metastasis was 13.3% (2/15). In conparison of the positive rates between these two groups, there was significant difference (χ 2=28.42, 24.67, P<0.001); but there were insignificant differences among cancers in different Duke’s staging group, different age group, different parts and categories (P>0.05). In comparison of 5～year survival rate in patients with positive expression of MDM 2 gene protein to that in patients with negative one, there was significant difference (χ 2=6.40, P<0.05). Negative expression was found in lateral tissues of cancer of 30 samples and in benign lesion tissues of 16 samples.Conclusions MDM 2 gene played an important role in the courses of infiltration of carcinoma of large intestine and the lymphatic metastasis, so MDM 2 could be an important index to prognose and diagnose carcinoma of large intestine.更多还原
