脑缺血诱导大鼠海马神经细胞凋亡及其相关蛋白Cpp32基因表达的初步研究
Studies on cerebral ischemia induces neuron apoptosis and gene expression of correlative protein Cpp32 in rat hippocampus
【摘要】 目的 探讨急性全脑缺血损伤后大鼠海马神经细胞凋亡及相关蛋白Cpp3 2的基因表达、分布。方法 运用DNA梯形电泳、免疫印迹、免疫组化分别分析了全脑缺血后海马神经细胞损伤、Cpp3 2蛋白含量变化及其分布。结果 15min急性全脑缺血复灌 2 4h后 ,大鼠海马呈明显细胞凋亡特征 ,且天胱蛋白 (Cpp3 2 )基因被持续激活表达 ,尤其是锥体神经细胞表现最为明显。 结论 急性全脑缺血能诱导海马神经细胞凋亡及Cpp3 2蛋白基因表达 ,其中锥体神经细胞表现最显著
【Abstract】 Objective To determine nerve cell apoptosis and gene expression and distribution of correlative protein Cpp32 in rat hippocampus after transient cerebral ischemia.Methods Using DNA ladder,western blotting and immunohistochemistry,we studied that hippocampus neuron injury,changes of Cpp32 content and its distribution following transient ischemia respectivly.Results (1)After 24 hours following 15min global brain ischemia,there were conspicuous cell apoptosis features in rat hippocampus.(2)Cpp32 gene were activated and expressive continuously,especially in the hippocampal CA1 neurons.Conclusion Transient cerebral ischemia may induce hippocampus neuron apoptosis and gene expression of Cpp32,the hippocampal CA1 neurons are more obvious.
