转化生长因子β2(TGF-β2)对大鼠角膜损伤愈合的影响

【摘要】 目的:探讨转化生长因子β2(TGF-β2)对大鼠角膜针刺损伤愈合的影响。方法:雄性Wistar大鼠20只,均取右眼制造角膜针刺创伤模型后随机分成实验组和对照组,实验组用转化生长因子β2(TGF-β2)点眼,对照组用生理盐水点眼,每日6次,至创面愈合止。计算剩余损伤面积、行组织学检查及透射电镜观察。结果:术后两组创面愈合面积比较实验组均快于对照组,且差异显著,组织学检查及电镜观察均显示两组差异。结论:TGF-β2有促进角膜损伤愈合的作用,其可能的机制为TGF-β2是胶原溶酶的主要抑制剂,可抑制基质中胶原酶的合成,降低胶原分解,促进胶原的分泌。

【Abstract】 Objective:To determine the effects of TGF-β2 on corneal wound healing in rat. Methods: After 20 male Wistar rats were injured by needle puncture at their corneas of right eyes, they were divided randomly into experimental group and control group. TGF-β2 was topically administered in the experimental group and normal saline in the controls 6 times a day until corneal wounds were healing. We measured the healing areas and evaluated the results by histology and electron microscopy. Results: Healing in experimental group was markedly faster than in control,and histologic examination showed the difference between the two groups. Conclusion: TGF-β2 can accelerate the corneal wound healing. The potential mechanism is that TGF-β2 as a main inhibitor of collagen dissolve zyme can restrain the synthesis of collagenase in matrix and promote the secretion of collagens.
