骨髓间充质细胞外源基因的表达和向脂肪细胞的分化

【摘要】 目的 研究骨髓间充质细胞的体外长期培养下外源基因的表达和向脂肪细胞分化的能力。方法 常规方法培养骨髓间充质细胞 ,免疫细胞化学方法对未分化的细胞进行鉴定。N2培养基诱导细胞向脂肪细胞分化 ,采用Ad 5 βgal检查骨髓间充质细胞对外源基因的感染效率。结果 从成人大鼠的骨髓中分离出骨髓间充质细胞 ,在体外生长形态似成纤维细胞 ,可以维持在未分化状态稳定增殖 ,体外扩增可超过 10多代。细胞可分化为脂肪细胞。结论 骨髓间充质细胞可在体外稳定增殖 ,能够表达外源性的基因 ,可以做为基因治疗的一个载体 ,能够分化为脂肪细胞 ,有潜力成为外科整形治疗的自体移植的一种细胞来源

【Abstract】 Objective: To investigate the long-term culture conditions of bone marrow stromal cells, their efficacy of expressing exogenous gene and their ability to differentiate into adipocytes. Methods: Cells were cultured routinely, undifferentialed cells were identified with immunocytochemistry and were induced to differentiate into adipocytes by N2 medium. Ad5 B gal was applied to test their capacity of cepressing foreign gene. Results: A population of bone marrow stromal cells could be isolated from adult rat bone marrow. They were processed to obtain a fibroblast-like population of cells and could be maintained in vitro for extended periods with stable population doubling, and were expanded as undifferentiated cells in culture for more than 10 passages, and they could be differentiated into adipocytes by N2 medium. Conclusions: Bone marrow stromal cells could be expanded in vitro stably. They were capable of expressing foreign gene and were the ideal cell sources for gene therapy. They could differentiate and adipocytes and represent an altemative autologous stem cell source for plastic surgery.
