过氧亚硝基阴离子在脑缺血再灌注损伤中的作用
Effect of nitric oxide and peroxynitrite on cerebral ischemia-reperfusion injury
【摘要】 目的 探讨过氧亚硝基阴离子 (ONOO- )在家兔脑缺血再灌注损伤中的作用。方法 采用三血管阻断法复制家兔脑缺血再灌注模型 ,分别测定对照组、单纯缺血组、再灌注损伤组脑组织匀浆内超氧化物歧化酶 (SOD)、丙二醛 (MDA)、NO- 2 /NO- 3(代表一氧化氮水平 )、诱导型一氧化氮合酶 (iNOS)水平的变化。结果 缺血再灌注组的NO- 2 /NO- 3、iNOS、MDA水平明显高于其他两组 (P <0 .0 5 ,P <0 .0 1 ,P <0 .0 5) ,而SOD的活性显著降低 (P <0 .0 5)。结论 脑缺血再灌注时脑组织内有大量的ONOO- 产生 ,ONOO- 参与脂质过氧化 ,介导了脑缺血再灌注损伤
【Abstract】 Objective To study the effect of peroxynitrite anoin(ONOO -) on ischemia-reperfusion injury of the brain.Methods Ischemia-reperfusion injury of the brain in rabbits was duplicated by trivascular blockage method,and the contents of melondialdehyde(MDA),NO - 2/NO - 3,iNOS and the activities of superoxide dismutase(SOD) and in homogenate of the cerebral tissues were observed in the control group, ischemia group and the ischemia-reperfusion injury group(IR) respectively SOD in the brain tissues.Result Compared with the group of the ischemia and the control group,the contents of Inos,NO - 2/NO - 2,MDA in ischemia-reperfusion group increased significantly(P<0.01, P<0.05,P<0.01) and the activities of SOD decreased markedly(P<0.05) in this IR group.Conclusion ONOO - are involved in oxidant-mediated brain injury following ischemia-reperfusion.
