用EMS诱变和小孢子培养快速获得甘蓝型油菜高油酸种质材料的研究

【摘要】 用EMS(甲基磺酸乙酯)溶液诱变甘蓝型油菜的小孢子,离心去除EMS,用秋水仙碱溶液对诱变后的小孢子进行染色体加倍,再离心去除秋水仙碱,用NLN 13培养基进行小孢子培养。当小孢子发育成完整的试管植株后,用幼叶氧化法定性、快速地筛选高油酸突变材料。筛选获得的材料经移栽定植,用气相色谱法精确地分析其种子的脂肪酸组成,得到了一份油酸含量为80 3396%的高油酸突变材料。本研究提高了油酸改良工作中诱变频率,降低测试分析费用。

【Abstract】 The microspores of Brassica napus were treated with EMS(a mutagen named ethyl methane sulphonate),then centrifugelized to eliminate the EMS.Then,the chromosomes of treated microspores were doubled with colchicines,then centrifugalized to eliminate the colchicines.Finally,the microspores were cultured in NLN13 medium.After the microspores developed into plantlets,the oleic acid contents of these materials were qualitatively and rapidly determined with the method of young leaf oxidation.The selected germplasmic materials were transplanted in soil.Finally,the fatty acid composition of the seeds were analyzed by gas chromatography.As a result,we obtained a high oleic acid germplasm containing 803396?% of oleic acid.The frequency of the mutant was increased and the cost for analyzing the fatty acid was reduced.
