蚕豆茎疫病菌rep-PCR初析
Preliminary rep-PCR analysis of Psudomonas fabae of broad bean
【摘要】 对分离自云南省 18个县 (市 )的蚕豆细菌性茎疫病菌 34个菌株进行的rep PCR分析结果表明 ,用引物BOX和ERIC分别扩增出 2 5和 2 4条多态性条带 ,分子量大小从 10 0~ 2 12 2 6bp ,大多数条带主要集中在 5 6 4~ 1375bp。蚕豆茎疫病菌菌株具有丰富的DNA多态性和较大的遗传变异。 2 4~ 2 5个菌株在 75 %遗传水平上相似。研究发现来自景东、昭通、玉溪、宣威、弥渡、文山、洱源、保山的菌株具有 75 %的同源性 ,与菌株的地理来源有一定相关关系。
【Abstract】 rep-PCR analysis for thirty-four isolates of Psudomonas fabae collected from 18 broad bean-producing counties in Yunnan was conducted.The results showed that 25 and 24 polymorphism bands were amplified respectively by using primers BOX and ERIC.The molecular weight of clust bands ranged from 100 bp to 21226bp,and most of them were 564bp to 1375bp.Analysis of genetic variation showed abundant DNA polymorphism and diversity among isolates,24 or 25 isolates had 75?% genetic similarity.And strains collected from Jindong,Zhaotong,Yuxi,Xuanwei,Midu,Wenshan,Eryuan and Baoshan had 75?% homology,indicating that it was related to the geographical origin of the pathogen in a certain degree.
