异丙酚对兔脊髓缺血再灌注损伤时神经源性胞浆酶的影响
Effects of Propofol on Cytosolic Enzymes during Spinal Cord Ischemia Reperfusion in Rabbits
【摘要】 目的观察异丙酚对兔脊髓缺血再灌注损伤时血清神经源性胞浆酶的变化与影响,以探讨异丙酚的保护效果及其机制。方法24只家兔随机分为假手术组、缺血再灌注组和异丙酚组,肾下主动脉阻断40min后松开再灌注60min。异丙酚组于阻断主动脉前沿耳缘静脉注射异丙酚5mg·kg-1,继以20mg·kg-1·h-1微量泵输注40min,假手术组与缺血再灌注组以同样方法静脉注射等容积生理盐水。测定阻断前、缺血40min及恢复灌注后60min时血清CK、CK—BB、LDH、AST、MDA的含量及观察术后3天脊髓的形态学改变。结果异丙酚明显降低血清CK、CK—BB、MDA的含量和减轻脊髓的形态学改变。结论异丙酚能明显减轻脊髓缺血再灌注损伤时血清神经源性胞浆酶的升高,对主动脉阻断所致的脊髓损伤具有保护作用。
【Abstract】 Objective To observe the effect of propofol on the changes of serum cytosolic enzymes and MDA during spinal cord ischemia reperfusion in rabbits. Methods 16 rabbits were randomly divided into ischemia group (A,n=8) and propofol group (B,n=8).The infrarenal aorta were clamped for 40 minutes then undamped. In group B, propofol was given intravenous injection 5 mg·kg-1 before clamping and then infused at a rate of 20 mg·kg-1·h-1 for 40 minutes,0.9% saline was used as control in group A. The activities of cytosolic enzymes and the production of MDA before, during and after cross-clamping were assayed. After operation the morphology of spinal cord was observed. Results Propofol can significantly reduced the increase of the activities of CK,CK-BB and the production of MDA, protected the morphologic alteration of spinal cord. Conclusion Propofol can protect the spinal cord from ischemia reperfusion injury during infrarenal aortic cross-clamping.
