急性血管性异种排斥反应中心肌的病理变化与T细胞的免疫组织化学研究
Immunohistochemical Study for T-lymphocyte and Pathological Change of Cardiac Muscle During Acute Vascular Xenograft Rejection
【摘要】 目的用仓鼠对小鼠心脏移植模型 ,探讨T细胞在异种急性血管性排斥反应中的作用。方法建立仓鼠对小鼠颈部异位心脏移植模型。分三组 :仓鼠—Balb/c小鼠 ,仓鼠—Balb/c裸小鼠 ,仓鼠—裸小鼠 (移植术后第 1 0 0日自小鼠尾静脉输 3× 1 0 7Balb/c脾细胞 )。观察移植物的平均存活时间及病理学变化 ,移植心组织内CD4+及CD8+T细胞的浸润。结果各组移植心的平均存活时间分别为 3 .3 8± 0 .5 2、>1 0 0、8.3 3± 2 .1 4日 ;排斥的移植心病检提示为典型的异种急性血管性排斥反应 ,可见CD4+T细胞浸润 ;而移植于裸鼠的长期存活移植心结构完整 ,未见细胞浸润。结论排斥反应依赖于T细胞的存在 ,CD4+T细胞在介导该排斥反应中起重要作用
【Abstract】 Objective To examine the role of T cell mediate immune response in a acute vascular rejection(AVR) using a Syrian hamster to mouse heart transplantation model.Methods The model of cervical heterotopic cardiac transplantation was performed using a Syrian hamster to mouse xenograft.Experiment animals were divided into three groups:Syrian hamster to mouse,Syrian hamster to nude mouse,Syrian hamster to nude mouse(inject nude mice with 3×10 7Balb/c lien cells at the 100th day after transplantation).Observe the mean survival time (MST) and pathological change of cardiac muscle in xenografts.Observe CD4 + and CD8 + T lymphocyte of cardiac muscle in xenografts with immunohistochemistry methods.Results The cardiac xenografts survival time were 3.38±0.52,>100,8.33±2.14 days respectively.The rejected xenografts demonstrated AVR manifestation with CD4 +T infiltration.In contrast,nude mice demonstrated normal intact cardiac xenografts without any cellular infiltration.Conclusion T cell dependent xenogenetic immune response plays a key role in concordant xenotransplantation.CD4 +T cells are involved in acute vascular rejection.
