【摘要】 目的 :评价用自制的抗猪囊虫单克隆抗体建立I-ELISA检测方法应用于临床脑囊虫病的诊断价值。方法 :用低温无水乙醇沉淀法分离纯化单抗IgG(腹水抗体效价 1∶32 0 0 ) ;用改良的过碘酸钠法制备辣根过氧化物酶标单抗 ;用抑制性酶联免疫吸附法 (I-ELISA)检测患者脑脊液和血清标本中囊虫特异性抗体。结果 :检测 6 3例确诊为脑囊虫病的患者脑脊液 ,抗体阳性 6 1例 ,阳性率为 97% ;血清 6 0例 ,抗体阳性 5 3例 ,阳性率为 88% ;其他颅内感染疾病患者脑脊液标本 2 4 0例 ,血清标本 2 4 0例 ,肝肺包虫病患者血清 10例 ,正常脑脊液 2 5例 (阑尾炎手术腰麻患者 ) ,健康志愿者血清 32例 ,检测结果均为阴性。结论 :抗囊虫抗原的单克隆抗体用常规鼠源B细胞杂交瘤技术获得 ,该单克隆抗体具有相当高的特殊性和检出率 ,对诊断脑囊虫病有很高的临床应用价值更多还原

【Abstract】 Objective: By using I-ELISA methed modified by this article authors to evaluate the clinical practical application value of the monoclonal antibody(McAb) against cysticerus cellulosae antigen in diagnosing cerebrocysticercosis. Methods: The McAb (immunoglobulin G titre 1:3200 in ascites) was separated and purified by using anhydrous ethanol at low temperature; HRP conjngated McAb was prepared by modified sodium periodate method; the antibody to cysticerus cellulosae antigen in patient’s cerebrospinal fluid and serum was determined by inhibitory ELISA(I-ELISA) method. Results: Out of 63 cerebrospinal fluid specimens from patients with clinically diaginosed cerebrocysticerosis, 61 specimens showed the anti cystscerus cellulosae antibody to be positive;positive rate 97%;53 of 60 serum specimens showed the antibody to be positive, positive rate 88%; 240 cerbrospinal fluid specimens and 240 serum specimens from patients with non cerebrocysticercous intracranial infections,10 serum specimens from patients with hepatic and pulmonary echinococcosis,25 cerebrospinal fluid specimens from patients with appendicitis and 32 serum specimens from healthy volunteers totally showed the anti cysticerus antibody to be negative. Conclusion:The MCAb against cysticerus cellulosae antigen has been developed by using routinely B cell hybridoma technique,and is of high specificity and detectable rate,and therefore, is of great clinical practise value in diagnosing cerebrocysticercosis.更多还原
