【摘要】 目的 :制备抗猪鼻支原体 (M .hyorhinis,M .hy)的单抗隆抗体 (monoclonalantibody ,McAb) ,为相关研究提供工具。方法 :用灭活的猪鼻支原体免疫BALB/c小鼠 ,运用传统的细胞融合技术制备抗猪鼻支原体单克隆抗体。单抗鉴定采用酶联免疫吸附实验 (ELISA)和免疫组织化学 (immunohistochemistry ,IHC)方法。结果 :经ELISA筛选获得了一株抗猪鼻支原体的单克隆抗体 ,该抗体亚类为IgG2b。ELISA结果表明该单抗仅与猪鼻支原体反应而与肺炎支原体和发酵支原体不反应。用经PCR检测支原体阳性的胃癌切片做免疫组化 ,结果显示 ,该抗体能与PCR阳性的胃癌组织切片产生特异性反应。结论 :获得一株抗猪鼻支原体特异性单克隆抗体 ,可为相关研究提供工具。更多还原

【Abstract】 Objective:To develop and characterize monoclonal antibody (McAb) against M.hyorhinis (M.hy) for related research work. Methods: BALB/c mice were immunized with heat-inactivated M.hy and McAb against M.hy was prepared by using traditional hybridoma technique. The characterization of McAb was studied by enzyme-linked immunosorbent assay(ELISA) and immunohistochemistry (IHC). Results: One clone of hybridoma secreting McAb against M.hy was obtained. The immunoglobulin subclass of the McAb was identified as IgG2b. The McAb could specifically react with M.hy by ELISA and IHC. Conclusion: The authors established one hybridoma cell line (2G10) stably secreting specific McAb (2G10) against M. hy.更多还原
