热应激对血管内皮细胞DNA损伤效应的研究
Effects of heat stress on DNA damage of vascular endothelial cells
【摘要】 目的 :观察血管内皮细胞株ECV30 4在不同热应激条件下DNA受损情况。方法 :实验采用单细胞凝胶电泳法(SCGE)检测在不同热应激条件 ( 4 1℃ 2h、 4 3℃ 2h、 4 5℃ 2h)对ECV30 4细胞DNA的单链损伤情况。结果 :在不同的热应激条件下 ,固定热应激时间 ,随着热应激温度的逐步增加 ,采用SCGE法所观察到的受损细胞数显著增多 ,P <0 0 5 ,相应的慧尾长度显著增加 ,P <0 0 5。结论 :热应激对内皮细胞的DNA致伤肯定 ,可使ECV30 4细胞DNA发生不同程度的单链断裂。
【Abstract】 Objective:To observe the DNA damage induced by heat stress in vascular endothelial cell strain(ECV304).Methods:The cultured ECV304 cells were divided into control group and experiment groups.Two hours’heat stress with the temperature of 41℃,43℃,and 45℃ were performed on ECV304 cells of experiment group Ⅰ,group Ⅱand groupⅢ,respectively.Single cell gel electrophoresis(SCGE)was used to detect the DNA single-strand damage of ECV304 cells.Results:As the temperature of heat stress increased,the number of damaged ECV304 cells and the length of comet tails increased significantly.(P<0 05).Conclusion:Heat stress can give rise to DNA single-strand breaks of ECV304 cells.
