IL-6对TNF-α的细胞杀伤作用影响的实验研究

【摘要】 目的 :研究IL - 6在肿瘤细胞对TNF -α耐受中的作用。方法 :检测SMMC - 772 1细胞在TNF -α作用下IL - 6的分泌。同时应用MTT实验检测细胞存活率 ,观察IL - 6和阻断IL - 6活性对细胞存活率的影响。结果 :SMMC - 772 1细胞能分泌IL - 6 ,TNF -α能促进细胞分泌IL - 6。外源加入的IL - 6对SMMC - 772 1细胞生长起促进作用。但加入抗IL - 6抗体或抗IL - 6R抗体时 ,与对照组相比 ,SMMC - 772 1细胞对TNE -α的敏感性也显著增加 ,10ng/mLTNF -α对SMMC - 772 1细胞就有显著的杀伤作用 ,P <0 0 1。随着浓度的增加 ,细胞存活率显著降低。结论 :抗IL - 6抗体和抗IL- 6R抗体可以抑制SMMC - 772 1细胞生长 ,能明显增加SMMC - 772 1细胞对TNF -α的敏感性。IL - 6对SMMC -772 1细胞的生长起促进作用。

【Abstract】 Objective:To investigate the effect of IL-6 on the resistance of TNF-α to hepatic carcinoma cells.Methods:IL-6 secretion of SMMC-7721 cell line,induced by TNF-α,was observed.MTT was employed to determine the effect of IL-6 and anti-IL-6R on the survival ratio of SMMC-7721 cells.Results:SMCC-7721 can secrete IL-6 and TNF-α can promote its secretion.The addition of IL-6 resulted in a significant cell growth of SMMC-7721.Significantly enhanced sensitvity of SMMC-7721 to TNF-α was observed when interrupting the action of IL-6 to SMCC-7721 by simultaneously adding IL-6 and anti-IL-6 or anti-IL-6R antibody,which resulted in a remarkable decrease of the SMMC-7721 survival ratio.Conclusion:IL-6 can protect hepatic carcinoma cells against the cytotoxic effect of TNF-α and its neutralization can enhance the sensitivity of cancer cell to TNF-α,which indicates that IL-6 is a growth factor for SMMC-7721 cell lines.
