MMP-9及TIMP-1在内毒素诱导性葡萄膜炎大鼠中的表达
Expression of matrix metalloproteinase-9 and tissue inhibitor of metalloproteinase-1 in rat model with endotoxin induced uveitis
【摘要】 目的 探讨基质金属蛋白酶 9(MMP 9)及其组织型抑制剂 (tissueinhibitorofmetallopoteinase 1,TIMP 1)在内毒素诱导性葡萄膜炎 (endotoxininduceduveitis,EIU)模型中的表达及意义。方法 2 0 0 μg伤寒杆菌内毒素注射于SD大鼠双后足垫 ,建立EIU模型。在内毒素注射后的不同时间点处死大鼠 ,采用免疫组织化学方法和计算机图像分析系统检测MMP 9和TIMP 1在不同时间点的表达及其积分吸光度 (A)值。结果 内毒素注射 6h开始出现炎症 ,以后炎症加重 ,于 2 4h达到炎症高峰 ,以后炎症减轻 ,7d时基本消退。虹膜上皮细胞 ,睫状体上皮细胞和渗出的炎症细胞表达MMP 9和TIMP 1。MMP 9在 6h出现表达 ,18~ 2 4h达高峰 ,以后逐渐下降 ,7d消失。TIMP 1于 2 4h出现表达 ,72h达高峰 ,以后逐渐下降。结论 大鼠EIU模型炎症早期MMP 9活性增高 ,继而TIMP 1分泌增多 ,MMP 9活性受抑 ,炎症减轻。提示MMP 9可能是参与EIU的重要调控因子 ,而TIMP 1则在炎症的抑制过程中发挥了重要作用。
【Abstract】 Objective To study the expression and role of matrix metalloproteinase 9(MMP 9) and its tissue inhibitor(TIMP 1) in Sprague Dawley rats with endotoxin induced uveitis(EIU).Methods Endotoxin induced uveitis model was established by injecting a dose of 200μg endotoxin (LPS) into the footpads of SD rats. At different time points (0,6,12,18,24,48,72,96 hours and 7 days) after LPS injection, the expressions of MMP 9 and TIMP 1 were detected immunohistochemically, and the results were analyzed by computer medical image analysis system. Results Intraocular inflammation appeared at 6 hours after LPS injection, peaked at 24 hours, and was resolved by the 7th day. MMP 9 and TIMP 1 were expressed in the epithelial cells of ciliary body, iris and exudated inflammatory cells. The level of MMP 9 elevated at 6 hours, peaked at 18~24 hours, dropped gradually afterwards, then disappeared at the 7th day. TIMP 1 was expressed at 24 hours, reached peaks at 72 hours, and decreased gradually to the baseline level.Conclusion The activity of MMP 9 increased in the early stage of EIU in SD rats. Later TIMP 1 was secreted and increased to inhibit the inflammation by restraining the activity of MMP 9. It’s suggested that as a modulating factor, MMP 9 has taken an active part in the course of EIU, and TIMP 1 has played an important role in inhibiting the inflammation.
