弱视猫视皮质17区Fos蛋白表达及治疗后改变

【摘要】 目的 观察剥夺性弱视猫治疗前后视皮质 ( visual cortex,VC) 17区 Fos蛋白表达的变化 ,探讨弱视治疗前后其神经元功能状态变化与 Fos蛋白表达的关系。方法 幼猫4周龄时缝合右眼眼睑建立剥夺性弱视模型 ,并在 8周龄时打开右眼 ,缝合左眼进行遮盖治疗。应用免疫组织化学染色法观察治疗前后视皮质 17区 Fos蛋白免疫阳性细胞密度的变化。结果 同年龄段的剥夺组较正常对照组免疫阳性细胞减少 ( P<0 .0 5 )。治疗组较剥夺组 ( )免疫阳性细胞增加 ,但仍低于正常组 ( ) ( P<0 .0 5 )。结论 剥夺性弱视猫 VC17区 Fos蛋白表达水平降低 ,并可通过缝合对侧眼改善。Fos蛋白法对检测视皮质神经元的功能状态具有一定敏感性 ,可以成为衡量弱视 VC兴奋性状态与形态学定位相结合的实验方法

【Abstract】 Objective To study the relationship between the inducibility of Fos protein and the neural activity level in visual cortex(VC) of amblyopia cats. Methods Cats were sutured right eyes in 4 weeks old. Then opened right eyes and sutured left eyes in 8 weeks old. It was observed the density of Fos protein expression in zone 17 of VC by Fos immunohistochemical method. Results The density of Fos immunoreactive cells in normal kittens was higher than in deprived kittens at the same age. While reverse suture could improve the density of Fos immunoreactive cells. Conclusion Fos protein expression in zone 17 of VC changed in deprived amblyopia cats and improved after reverse suture. Fos immunohistochemical method is an experimental method, which can contact the activity of neural cells with its location in VC and evaluate the alterations of vision functions in amblyopia.
