阿霉素抑制兔眼晶状体上皮细胞增殖细胞核抗原表达的实验研究
Studies on adriamycin inhibiting expression of proliferating cell nuclear antigen in rabbit lens epithelial cells
【摘要】 目的 探讨人工晶状体植入术后阿霉素 (adriamycin,ADM)抑制兔眼晶状体上皮细胞 (rabbit lens epithelial cells,RL EC)增殖的作用及客观评价后囊膜混浊 (posteriorcapsule opacification,PCO)的方法。方法 采用兔眼晶状体囊外摘出联合人工晶状体植入术的动物模型 ,术中将携带 ADM药物缓释系统植入 15只兔眼晶状体囊袋内。术后观察后囊膜混浊情况 ;术后第 4周 ,链霉亲和素免疫组化法 (strept- avidin- biotin- enzyme com ple,SABC)检测兔眼晶状体上皮细胞的增殖细胞核抗原 (proliferating cell nuclear antigen,PC-NA )的表达。结果 实验组与对照组比较 ,兔眼晶状体上皮细胞的 PCNA阳性表达下降 ,2组比较差别具有显著性意义。结论 ADM能有效抑制人工晶状体植入术后 RL EC的增殖 ,免疫组化技术 (SABC法 )检测 RL EC的 PCNA的表达 ,为实验室客观评价 PCO提供一种简便有效的方法
【Abstract】 Objective To investigate adriamycin (ADM) inhibiting rabbit lens epithelial cell(RLEC) proliferation and objective means for evalluating posterior capsule opacification (PCO) after intraocular lens(IOL) implanting operation.Methods A PLA disk containing 22μg ADM was implanted between the posterior capsule and IOL during the extracapsular cataract extraction(ECCE) with IOL implanting operation. In fifteen rabbits eyes at 4 weeks after operation, RLEC were examined by immunohistochemical technique (strept avidin biotin enzyme comple, SABC) for identifying the expression of proliferating cell nuclear antigen (PCNA). Results The level of PCNA in test group was significantly lower than that in control group at 4 weeks after operation.Conclusion ADM can prevent proliferation of RLEC and immunohistochemical technique can provide an efficient, facilitate and objective means for valuing PCO after IOL implanting operation.
