IVF实验室质控中影响鼠胚体外培养的因素分析

【摘要】 目的 :建立适宜的小鼠胚胎体外培养体系 ,探讨CO2 气体纯度和培养器皿对小鼠胚胎体外发育的影响。 方法 :采用微滴法培养 2 细胞鼠胚 ,以 4~ 8 细胞鼠胚和桑椹胚形成率作为判断培养效果的指标。 结果 :普通纯度CO2 气体培养组的 4~ 8 细胞形成率、桑椹胚形成率分别为 12 .5 %、0 .0 % ,高纯度CO2 气体培养组分别为6 7.6 %、4 4 .1% ,两组比较差异有统计学意义 (P均 <0 .0 0 1)。国产玻璃器皿培养组的 4~ 8 细胞形成率、桑椹胚形成率分别为 74 .4 %、5 3.5 % ,进口一次性器皿培养组分别为 85 .7%、6 2 .8% ,两组比较差异无统计学意义 (P >0 .0 5 )。结论 :高纯度CO2 气体更适宜鼠胚的体外发育 ;国产玻璃器皿与进口一次性器皿对鼠胚体外发育的效果相近

【Abstract】 Objective: To establish more suitable in vitro culture system of embryos, studied the effects of purity of CO\-2gas and different types of culture dishes on in vitro development of mouse embryos. Methods: 2 cell mouse embryos were cultured in microdrops of medium and the percentage of embryos developed to 4~8 cell embryos and morular stage was used as evaluated criteria. Results: (1) There were significantly higher 4~8 cell embryos, morula percentages in high purity CO\-2gas group (67.6%, 44.1% respectively) as compared those in common purity CO\-2gas group (12.5%, 0.0% respectively). (2) There were no significant difference 4~8 cell embryos, morula percentages between the homemade culture dishes group (74.4%, 53.5% respectively) and the imported disposal culture dishes group (85.7%, 62.8% respectively). Conclusions: High purity CO\-2gas was more suitable in supporting in vitro development for mouse embryos. Both the homemade and the imported disposal culture dishes produced good results of embryos in vitro development.
