食管癌3p24等位基因LOH及其扩增产物克隆的研究
Loss of heterozygosity and cloning of human chromosome 3p24 in esophageal carcinoma
【摘要】 目的:研究 3p2 4 EAβMD位点与食管癌的关系 ,为寻找该位点附近可能存在的抑癌基因奠定基础。 方法 :采用 PCR- RFL P法检测 4 5例食管癌患者 3p2 4位点杂合缺失情况 ,并对该片段进行克隆。结果:在 2 2例食管癌信息个体中共检出 9例杂合缺失 ,并通过序列分析可知变化为点突变。结论 :3p2 4 EAβMD较高的杂合缺失率显示该位点附近可能存在潜在的抑癌基因
【Abstract】 Objective: To investigate the association between the loss of heterozygosity on chromosome 3p24 and the prevalence of esophageal carcinoma. Methods: The tissue DNA was extracted from esophageal carcinoma and normal tissue. The loss of heterozygosity on chromosome 3p24 was conducted by means of polymerase chain reaction restrict fragment length polymorphisms (PCR RFLP). With gene cloning, two differential alleles were successfully cloned and analyzed. Rusult: 45 pairs of specimens obtained from patients with esophageal carcinoma were tested by PCR RFLP. On EAβMD site of 3p24, the LOH was detected 9 of 22. The point mutation was observed in this site. Conclusion: It suggests that the LOH on EaβMD site of 3p24 may probably correlate with the prevalence of esophageal carcinoma.
