过量氟对体外培养小鼠磨牙牙胚发育的影响
The effects of fluoride on the development of mouse molars in vitro
【摘要】 目的:建立小鼠牙胚发育的体外培养模型 ,初步研究过量氟对小鼠牙胚发育的影响。方法 :取胎龄 18dBAL B/ c小鼠牙胚 ,在 96孔培养板 BGJb半固体培养基标准环境下培养 5 d和 10 d,HE染色 ,观察牙胚发育形态 ,并初步研究 5 .0× 10 - 6 g/ L 氟对体外牙胚发育的影响。结果:牙胚在体外继续发育 ,牙尖形成明显 ,内釉上皮细胞和成牙本质细胞极化 ,分泌基质。氟干预组牙胚发育与对照组相似 ,但部分区域成牙本质细胞极化不明显 ,基质分泌明显少于对照组。 结论 :小鼠牙胚在 BGJb半固体培养基生长良好 ,为研究氟对牙胚损伤提供良好模型。
【Abstract】 Objective: To establish the model of the cultured tooth germ in vitro and examine the effect of fluoride on it. Methods: The tooth germs from embryonic 18th d mice’s first maxillary molar were devided into 2 groups; the control group and the experiment group. The explants were cultured in semi solid BGjb medium and the experiment group contained 5.0×10 -6 g/L fluoride. After cultured for 5 days and 10 days, HE staining were used. Results: It showed that two groups of tooth germs developed well in vitro. The cusps were evident. The inner enamel epithelia and odontoblasts were polarized and secreted matrix, while some of the odontoblasts polarized not very well and the matrix was relatively thinner in the fluoride group. Conclusions: Mice’s tooth germs developed well in semi solid BGjb medium. It can provide the model for studying the effects of fluoride on the tooth germ development.
