亚砷酸钠对人皮肤细胞增殖作用的影响
Effects of proliferation on primary cultured human skin cells induced by sodium arsenite
【摘要】 目的:探讨亚砷酸钠对人皮肤细胞增殖作用的影响。方法:使用原代培养的人真皮成纤维细胞和表皮细胞 ,采用直接细胞计数法和 MTT法检测亚砷酸钠对两种细胞系生长的影响。 结果:成纤维细胞较表皮细胞易于培养 ;MTT法检测 ,不同剂量的亚砷酸钠与对照组比较 ,吸光度值均有提高 ,有剂量 -效应关系 (P <0 .0 1) ;0 .5~8.0 μm ol/ L 的亚砷酸钠对成纤维细胞有明显增殖作用 ;低浓度 (0 .8μmol/ L、1.6 μm ol/ L)对表皮细胞有明显的增殖作用 ,浓度高于 3.2 μm ol/ L 时 ,表皮细胞生长受抑制 ;10 .0 0 μmol/ L 亚砷酸钠对成纤维细胞有毒性作用。 结论 :无机砷诱导人皮肤细胞的增殖作用可能是引起慢性砷中毒皮肤损伤的重要原因 ,具体作用机制有待进一步研究
【Abstract】 Objective: To investigate the effects of injury on primary cultured skin cells induced by sodium arsenite. Methods: Count cell number, MTT reduction assay,transmission electron microscope were performed in primary cultured skin cells, sodium arsenite was used as tester. Results: With different concentrations of 0.5~8 μmol/L sodium arsenite, the skin fibroblasts were markedly stimulated and showed a dose effect relationship. Sodium arsenite at lower concentrations (0.8 μmol/L?1.6 μmol/L) induced mainly the degree of proliferation of epidermic cell, while inhibiting cell growth over 3.2 μmol/L. At concentration of 10 μmol/L, however, sodium arsenite induced injury of skin fibroblasts. Conclusion: The proliferation of skin cell may play a critical role in the development of skin injury caused by inorganic arsenic, the mechanism of DNA injury in primary cultured human skin cells needs further study.
