红毛藻藻红蛋白的粗提方法比较及不同光照条件下藻胆蛋白变性机制的初步探讨

【摘要】 比较研究了多种细胞破碎方法和盐析方法对红毛藻中藻红蛋白的提取效果,确定组织捣碎法为效率较高的细胞破碎方法,用饱和度为60%的硫酸铵按"改进结晶法"进行盐析为较佳的盐析方法.探讨了冻融法的作用机制.同时对藻红蛋白盐析液在不同照射条件下的变性机制进行了初步的研究和探讨,研究显示光照有可能使藻胆蛋白的摩尔消光系数发生变化.

【Abstract】 Different methods of cell fragmentation and salting out were compared in the present study to obtain the optimal method for extraction of phycoerythin from the redalga Bangia Fuscopururea. The results showed that tissuetrituration method was a relatively appropriate method for cell fragmentation compared with other methods, and the improved crystallization method with 60% saturation degree of ammonium sulfate was a more efficient salting out method than any other method used. The mechanism of freezing and thawing method was also discussed. The possible denaturalization mechanism of phycobiliprotein in salting out solution under different kinds of irradiation was experimentally studied. It was suggested that the irradiation could change the molar extinction coefficients of phycobiliprotein and caused the denaturalization of phycobiliprotein.
