高效液相色谱法测定格拉丝琼血药浓度
Measurement of granisetron in plasma by high performance liquid chromatography
【摘要】 目的建立格拉丝琼血药浓度的高效液相色谱测定方法。方法以甲醇-0.05mol·L-1醋酸钠缓冲液(pH6.0,含0.25%三乙胺)(97:3)为流动相,激发波长为305nm,发射波长365nm,血浆样品采用乙酸乙酯提取后进样。结果格拉丝琼在0.17.5μg·L-1浓度范围内线性关系良好,平均回收率(99.5±8.7)%,日内RSD≤8.0%,日间RSD≤9.7%。结论该方法简便,准确,可用于格拉丝琼临床药动学研究。
【Abstract】 OBJECTIVE To establish a HPLC method to measure the plasma concentration of granisetron.METHOD Mobile phase was a mixture of methanol and 0.05 mol · L-1 sodium acetate buffer(pH6.0) contained 0.25%(v/v) triethylamineater(97:3).A fluorescence detector of 305 nm for excitation and 365 nm for emission was used.Ethyl acetate was used for liquid-liquid extractioa RESULTS The assay procedure was shown to produce linear calibration curves over the range 0.1μg · L-1 to 7.5 μg · L-1.Within the range,the recovery rate was(99.5±8.7)%.The coefficients of variation within a day and between days did not exceed 8.0%,9.7%,respectively.CONCLUSION This method is found to be easy,convenient and precise for clinical pharmacokinetics study of garnisetron.
