白细胞介素-6对周围神经损伤后神经细胞FOS表达的影响(英)

【摘要】 背景:神经系统正常情况下低水平FOS表达,损伤后FOS表达增强,白细胞介素-6(interleulin-6,IL-6),在中枢神经系统损伤后能抑制,神经细胞FOS表达,但周围神经损伤后神经细胞FOS表达与IL-6的关系的研究目前尚未见报道。目的:观察周围神经损伤后神经细胞FOS表达及IL-6对其表达的影响。设计:完全随机对照的前瞻性实验研究。地点和对象:2002/08~12在广东医学院实验动物中心完成,实验对象为DS大鼠48只,体质量250~340g,雌雄不拘。干预:大鼠在无菌条件下显露左侧坐骨神经,实验组神经内注射IL-62mL,对照组神经内注射生理盐水2mL。术后1h挤压损伤坐骨神经,分别于伤后1,2,3,4h取出标本进行免疫组化染色。主要观察指标:观察实验组和对照组FOS免疫阳性细胞。结果:伤后1,2,3,4h对照组FOS免疫阳性细胞分别为548±14,640±12,718±21,732±11;实验组分别为202±13,212±17,245±22,288±14。随挤压时间的延长而增加。实验组各时间FOS免疫阳性细胞数较对照组明显减少(t=-1.812,2.128,P<0.05;t=-2.864,3.159,P<0.01)差异有显著性意义。结论:IL-6对周围神经损伤后神经细胞FOS表达有抑制作用,提示IL-6参与神经损伤后的调控过程。

【Abstract】 BACKGROUND:Low level FOS expressions of neural system under normal circumstance increase after injury.Interleulin 6(IL 6)can inhibit neurocyte FOS expressions after the injury of central neural system;however,there is no report on the relationship between IL 6 and neurocyte FOS expressions after peripheral nerve injury. OBJECTIVE:To observe the expression of neurocyte FOS as well as the effect of IL 6 on neurocyte FOS expression after peripheral nerve injury. DESIGN:Forty eight SD rats were randomly divided into two group. SETTINGS AND PARTICIPANTS:The research was completed in Guangdong Medical College experimental animal center from August to December 2002.Objects:48 SD rat with a weight between 250 g and 340 g, either male or female were selected for the research. INTERVENTIONS:The left sciatic nerve of rat was exposed under asepsis condition;2 ml IL 6 was injected into the nerve of intervention group as well as 2 ml saline was injected into the same site of control group. The sciatic nerve was injured by extrusion 1 hour after the surgery, and the nerve was taken for immunohistochemical staining at 1,2,3,4 hours respectively after the injury. MAIN OUTCOME MEASURE: The FOS immune reaction(FOS IR) positive cells were observed in both intervention and control group. RESULTS:The FOS IR positive cells in control group at 1,2,3,4 hours after injury were(548±14, 640±12),(718±21),(732±11)as well as that of the intervention group were(202±13), (212±17),(245±22)and(288±14) respectively,which the values were increased along with the prolongation of the extrusion time.The FOS IR positive cells in intervention group at different time were significantly less than those of the control group(t=-1.812, 2.128,P< 0.05;t=-2.864, 3.159,P< 0.01). CONCLUSIONS:IL 6 has an inhibitory action on neurocyte FOS expressions after peripheral nerve injury, which suggests that IL 6 participates in the control process after nerve injury.
